Abstract

The aim of this study was to obtain in vitro haploid embryos within the Kahramanmaraş red peppers (Capsicum annuum L.) via androgenesis. The anthers, at the stage of uninucleate microspore excised from the unopened buds, placed onto the MS media and kept under the certain temperature and light. In order to induct direct embryogenesis through the anthers, a range of oksin (2.0, 4.0, 6.0 mg/1 NAA and 1.0, 2.0, 3.0, 4.0 mg/1 2,4-D) and cytokinin (0.1, 1.0, 2.0, 3.0 mg/1 BAP and 0.1, 1.0, 5.0 mg/1 kinetin) were added. Activated charcoal (0.25 %) and AgNOj (10 mg/1) were also included in the media. The following three different temperature pre-treatments were applied to cultured anthers for a week: control, 4 °C, 29 °C and 35 °C (without illumination). During the course of this research, a total of 37 treatments including different concentrations of oksin-cytokinin, activated charcoal and temperature pre-treatments were applied. A total of 9750 anters were cultured and some did produce callus. The anthers (327 in number) cultured on a media composed of MS+0. lmg/1 BAP +4mg/l NAA + 0.2 % aktivated charcoal + 10 mg AgNOs did not produce callus; however 9 embryos were developed from 3 anthers. With this media, the embryo/anther rate was 2.8 %.

Özet

Kahramanmaraş kırmız biberlerinde (Capsicum annuum L.) yürütülen bu çalışmada androgenesis yoluyla in vitro haploid embriyo oluşturma amaçlanmıştır.
