Abstract

Objective: Pyruvate is known as a key product of glycolysis, and it has antioxidant and anti-inflammatory effects. In the cell culture, sodium pyruvate is frequently used as an additional energy source. However, the concentration-dependent effects on human fibroblast cell behavior have not been clearly investigated. Methods: In this study, the effect of sodium pyruvate supplementation on the cell viability, spheroid formation capacity, and CCND1 gene expression in CCD-1072Sk human fibroblast cells was analyzed through MTT, spheroid formation assay and real-time qRT-PCR, respectively. Results: Sodium pyruvate treatment significantly affected the cell viability and spheroid formation in a dose-dependent manner. Furthermore, CCND1 gene expression was upregulated at concentrations of 2mM sodium pyruvate and above. Conclusion: These results suggest that sodium pyruvate may promote cell proliferation and spheroid formation, while modulating gene expression in human fibroblasts. Further studies are required to clarify the underlying molecular mechanisms and to better understand the broader implications of pyruvate supplementation in cell culture systems and potential biomedical applications.
