Abstract

Background: It has been shown that pathogenic plaque, diet, saliva, immune system, education level, oral hygiene habits as well as genetic factors may be effective in the formation of caries. Methylenetetrahydrofolate reductase (MTHFR) encodes enzymes involved in homocysteine metabolism. Polymorphisms in this gene significantly impact in folic acid and vitamin B12 metabolism. In our study, we investigated the genetic impact on dental caries by examining the association between the MTHFR rs1801133 polimorphism and DMFT (Decayed/Missing/Filled Teeth) in caries risk groups. Methods: 2 groups were formed consisting of 56 individuals: no caries (NC- control group, DMFT=0, n=28) and individuals with caries lesions (CL-study group, DMFT≥14, n=28) homogenized in terms of periodontal status. Genotyping was conducted using Real-Time PCR and a TaqMan kit with genomic DNA from participants. Genotype and allele distributions were compared using the χ² test, with statistical significance set at p<0.05. Results: In the CL-group; 15 had CC (53.6%), 8 had CT (28.6%), and 5 had TT (17.8%) genotype. In the NC-group;15 had CC (53.6%), 11 had CT (39.3%), and 2 had TT (7.1%) genotype. There was no significant difference in genotype and allelic distribution between CL and NC groups (p=0.4149 for genotype, p=0.5341 for allele). Conclusion: Analysis of genotype distribution revealed that the TT genotype was present in 17.8% of the CL-group and 7.1% of the NC-group. This proportional difference in TT genotype associated with lower folate levels may indicate that the MTHFR rs1801133 gene polymorphism may be associated with caries risk.

Bulgular: CL grubunda olan 28 bireyin 15'i CC (%53,6), 8'i CT (%28,6) ve 5'i TT (%17,8) genotipe sahip olmakla birlikte; NC grubunda olan 28 bireyin 15'i CC (%53,6), 11'i CT (%39,3) ve 2'si TT (%7,1) genotipe sahip olarak bulunmuştur.
