Abstract
This study aimed to evaluate the effects of a time-restricted feeding model on inflammasome-related gene expression associated with colonic inflammation. Twelve-month-old male Wistar rats were assigned either to a control group (ad libitum feeding) or to an intermittent fasting protocol consisting of 18 hours of daily fasting followed by 6 hours of feeding for 35 days (n = 7 per group). At the end of the experimental period, total RNA was isolated from colon tissues, and following cDNA synthesis, the expression levels of core inflammasome components (Nlrp3, Asc, and Caspase-1) and pro-inflammatory cytokine genes (Il-1β and Il-18) were analyzed using quantitative real-time PCR. Gene expression levels were statistically evaluated based on normalized ΔCt values. The intermittent fasting group exhibited significant transcriptional downregulation of all target genes compared to controls (p < 0.05). These findings indicate that time-restricted feeding may suppress colonic inflammatory responses by modulating the Nlrp3 inflammasome axis at both upstream (sensor/adaptor) and downstream (effector/cytokine) levels. Collectively, the results suggest that intermittent fasting may represent a complementary and preventive strategy in inflammation-based intestinal pathologies. Nevertheless, the molecular findings reported here require further validation through functional and clinical investigations.
Özet
Bu çalışma, zaman kısıtlamalı beslenme modelinin kolonik inflamasyonla ilişkili inflammasom gen ekspresyonu üzerindeki etkilerini değerlendirmeyi amaçlamıştır. On iki aylık erkek Wistar sıçanlar 35 gün süreyle kontrol (ad libitum beslenme) veya günlük 18 saat açlık–6 saat beslenme protokolünü içeren aralıklı açlık uygulamasına tabi tutulmuştur (n=7/grup).
