Abstract

In this study, seed and pollen morphology of the endemic Sabulina mesogitana subsp. flaccida, which naturally occurs in Turkey, was examined by light and electron microscopy. The objectives included in vitro germination of seeds, assessment of pollen viability, observation of pollen tube formation, and investigation of seed and pollen micromorphology. Seeds of S. mesogitana subsp. flaccida, collected from the Western Taurus Mountains, including the Küre Mountains National Park, one of the nine forest priority hotspots for conservation in Turkey, along with its buffer zone, were germinated in vitro using Murashige and Skoog (MS) media prepared with different sucrose concentrations and pH. The highest germination rate was observed at 34.3% in the medium containing 20 g/L sucrose and pH 5.8. To determine pollen viability, pollen tube formation, and pollen tube length, different sucrose concentrations were tested; the highest germination rate achieved was 30% in the medium containing 20 g/L sucrose. Additionally, the pollen fertility rate of the species was determined to be approximately 88%. Seed and pollen surface morphology was evaluated by scanning electron microscopy (SEM); seed surfaces on ventral and dorsal sides were found to be papillate and tuberculate, with dark brown to black coloration, while pollen grains exhibited a spherical shape with micro-echinate and micro-perforate ornamentation. Since morphological characteristics provide important data for taxonomic classification, this study presents seed and pollen germination data for the endemic S. mesogitana subsp. flaccida species.
