Afyonkarahisar Yöresi Koyun Akciğer Kılkurdu Türlerinin Moleküler Karakterizasyonu

Özet
Bu araştırma, Afyonkarahisar yöresinde koyunlarda akciğer kılkurdu enfeksiyonlarının moleküler epidemiyolojisi ve akciğer kılkurdu türlerinin moleküler karakterizasyonunu ortaya koymak amacıyla yapılmıştır. Bu amaçla 5 farklı lokalitede akciğer kılkurtları ile doğal enfekte 10 sürüden akciğer kılkurdu birinci dönem larvalarını izole etmek için dışkı örnekleri toplandı. Baermann-Wetzel yöntemi ile separe edilen akciğer kılkurdu 1. dönem larvalarının spin kolon yöntemiyle ticari kit (Thermo) kullanılarak DNA’ları çıkarıldı. PCR’da akciğer kıl kurtlarının 28S rRNA bölgelerine ait primerler (Dictyocaulus filaria: F-gctacaaaatcgcatacgaacg, R-actccttagcggttaccga; Cystocaulus ocreatus: F-cggtttgtttagcatgtccaaagtc, R-tctctgacttcgtcctgc; Muellerius capillaris: F-aaaggcccaacgctgaa, R-cctctgacttcgtcctgc) FastPCR programı ile dizayn edilerek kullanıldı. Toplam 7 izolattan elde edilen DNA PCR ürünlerinin 28S rRNA gen bölgelerinin DNA dizileme analizi yapıldı. Bir ve 2. izolatlardan elde edilen 952 bp uzunluğundaki dizilerinin %100 D. filaria, 4, 5 ve 6. izolatlardan elde edilen768 bp uzunluğundaki dizilerinin %100 C. ocreatus, 7. izolattan elde edilen 780 bp uzunluğundaki dizinin %100 M. capillaris ve 3. izolattan elde edilen 780 bp uzunluğundaki dizinin ise %99 ihtimalle M. capillaris olduğu görülmüştür. İzolat 7'nin 702. bazının heterozigot yapıda olması ve izolat 3'te 117 ve 718. bazlarında mutasyon olması bir alt tür olabileceğini düşündürmüştür. Bir sürüde rastlanılan Neostrongylus linearis larvasından DNA elde edilememiş, Sürülerde Protostrongylus rufescens larvasına rastlanmamıştır.
Abstract
The present study was conducted to investigate molecular epidemiology of lungworm infections and molecular characterization of lungworm species of sheep in Afyonkarahisar region. Fecal samples were gathered from 10 flocks in 5 different localization to isolate first stage larvae of lungworm. First stage larvae of lungworms were seperated with Baermann-Wetzel method and DNAs were extracted with spin-column method by using commercial kit (Thermo). Primers belonged to 28S rRNA region of lungworms (Dictyocaulus filaria: F-gctacaaaatcgcatacgaacg, R-actccttagcggttaccga; Cystocaulus ocreatus: F-cggtttgtttagcatgtccaaagtc, R-tctctgacttcgtcctgc; Muellerius capillaris: F-aaaggcccaacgctgaa, R-cctctgacttcgtcctgc) were used with the design of FastPCR software in PCR. DNA sequence analysis was perfomed on PCR products of 28S rRNA gene regions obtained from seven isolates. 952 bp length sequences of first and second isolates were D. filaria with the probability of 100%; 768 bp length sequences of 4th, 5th and 6th isolates were C. ocreatus with the probability of 100%; 780 bp length sequence of 7th isolate was M. capillaris with the probability of 100% and 780 bp length sequence of 3rd isolate was M. capillaris with the probability of 99%. Heterozygote structure of base number 702 of 7th isolate and presence of mutations in base numbers 117 and 718 of 3rd isolate suggest probability of a subspecies. DNA of Neostrongylus linearis larvae which was detected in one flock could not be obtained. Protostrongylus rufescens larvae was not found in flocks.
