Sıçanlarda Parasetamolle İndüklenen Karaciğer Toksisitesi Üzerine Berberis Vulgaris Bitki Ekstresinin Koruyucu Etkileri

Özet

Amaç: Berberis vulgaris (kızamık, karamut, adi kadıntuzluğu) bitki ekstresinin hem antiiflamatuvar hem de antioksidan özelliğinin olmasından dolay ı birçok hasara karşı koruyucu olduğu bilinmektedir. Parasetamol zehirlenmesi en s ık görülen zehirlenmelerden biridir. Çalışmamızda parasetamol toksisitesinin vermiş olduğu zararı Berberis vulgaris bitki ekstresinin yararlı olan etkileriyle azaltmayı planladık. Gereç ve Yöntem: Çalışmamızda 42 adet Sprague Dawley cinsi 8 haftal ık s ıçan kullanıldı. Her grupta 6 di şi hayvan olacak şekilde 7 grup oluşturuldu. Çalışmanın 24. saatinde tüm gruplara anestezi altıda laparotomi uygulandı ve karaciğer diseksiyonu yapıldı. Karaciğer histopatolojisini değerlendirmeler için Hematoksilen ve Eozin (H&E) bayama, karaciğer dokusunda glikojen ve bazal membran boyanmalarını göstermek amac ıyla PAS boyası yapıldı. Ayrıca TUNEL boyasıyla apoptotik hücreler gösterilerek skorlama yapıldı. Bulgular: Histopatolojik sonuçlarda parasetamol uyguladığımız grupta, hepatik dejenerasyon ve peritübüler inflamasyon oldukça belirgindi. Bu çal ışmada TUNEL boyamasında parasetamol verdiğimiz gurupta (Grup 4) pozitif hücre sayısının artt ığını gözlemledik. Yaptığımız PAS boyamas ı sonucunda parasetamolle indüklediğimiz grubun hepatositlerinde glikojen birikimi dikkat çekmekteydi. Sonuç: Parasetamol toksisitesi olan gruba yüksek doz Berberis vulgaris bitki ekstresi verildiğinde apoptotik hücre say ısının ve inflamasyonun azaldığı görüldü.

Abstract

Introduction: Berberis vulgaris extract is known to be protective against many damages due to its anti-inflamatory and antioxidant properties. Paracetamol toxicity is one of the most common toxicities. In our study, we aimed to reduce the damage caused by paracetamol toxicity with the beneficial effects of Berberis vullgaris extract. Material and Methods: In our study, 42 Sprague Dawley 8- week-old rats were used. 7 groups were formed with 6 female animals in each group. At the 24th hour of the study, all groups underwent laparotomy under anesthesia and liver dissection was performed. Hematoxylin and Eosin (H&E) staining was used to evaluate liver histopathology, and PAS staining was used to show glycogen and basement membrane staining in liver tissue. In addition, scoring was done by showing apoptotic cells with TUNEL stain. Result: Hepatic degeneration and pe ritubular inflammation were quite evident in the groups to which we applied paracetamol. In this study, we observed that the number of positive cells increased in the group given paracetamol (Group 4) in TUNEL staining. As a re sult of the PAS staining we performed, glycogen accumulation in the hepatocytes of the group we induced with paracetamol was remarkable. Conclusion: It was observed that the number of apoptotic cells and inflammation decreased in the paracetamol toxicity group given high dose Berberis vulgaris extract.
