O-Vanilin İçeren Schiff Bazı Ligandı ve Pd(II) Kompleksinin Glioblastoma Multiforme Hücrelerindeki Tedavi Potansiyelinin İncelenmesi ve Elektroporasyonun Etkinliği

Özet
Giriş: Glioblastoma multiforme (GBM) sık görülen ve en ölümcül primer beyin tümörlerindendir. Cerrahi rezeksiyon, radyasyon ve yüksek-doz kemoterapi gibi standart tedavilere rağmen, tan ı konulduktan sonra hastaların ortalama sağ kalım süresi on iki ay civarındadır. Amaç: Bu çalışmada, Metil 2-Amino-6-Metil-4,5,6,7- Tetrahidrotieno[2,3-c] Pi ridin-3-Karboksilat bazl ı ligand ve Pd(II) kompleksinin kemoterapiye dirençli GBM hücrelerindeki tedavi potansiyellerini araştırmak ve elektroporasyonun (EP) bu bileşiklerin sitotoksisiteleri üzerindeki etkinliğinin araştırılması amaçlanmıştır. Gereç ve Yöntem : Ligand ve Pd(II) kompleksinin sitotoksik aktivitesi T98G insan GBM hücre hatt ına kar şı araştırılırken, biyouyumluluk için sağlıklı fibroblast hücreleri (L-929) kullanıldı. MTT testi kullanılarak Ligand ve Pd(II) kompleksinin tek ba şına veya EP ile kombinasyon halindeki sitotoksisiteleri belirlendi. Bulgular: Pd(II) kompleksi T98G hücrelerinde iyi düzeyde anti- kanser aktivite gösterirken (IC 50= 394.67), L-929 fibroblast hücrelerinde sitotoksik aktivite göstermemiştir (IC50= 1449.28). Ligand ise T98G ve L-929 hücre hatlar ında s ırasıyla IC 50= 1126.79 ve IC50= 1283,36 düzeyinde sitotoksik aktivite göstermiştir. T98G GBM tedavisinde Pd(II) kompleksinin EP ile kombine uygulanması tek başına Pd(II) kompleksin uygulanmasından çok daha etkili olduğu tespit edilmiştir. Sonuç: Sağlıklı hücrelerde (L-929) sitotoksik aktivite göstermeyip, T98G hücre hatt ında sitotoksik aktivite gösteren Pd(II) kompleksi GBM tedavisi için olası kemoterapötik ajan olarak geliştirilebilir ve EP ile kombine kullanılması durumunda umut vaat eden sonuçlar verebilir.
Abstract
Introduction: Glioblastoma multiforme (GBM) is one of the most common and deadliest pr imary brain tumors. Despite standard treatments such as surg ical resection, radiation, and high-dose chemotherapy, the average survival time of patients after diagnosis is around twelve months. Objective: The aim of this study was to investigate the chemotherapy potential of Meth yl 2-Amino-6-Methyl-4,5,6,7- Tetrahydrothieno[2,3-c] Pyridine-3-Carboxylate-based ligand and its Pd(II) complex in chemotherapy-resistant GBM cells and to investigate the efficacy of electroporation (EP) on the cytotoxicity of these compounds. Material and Method: The cytotoxic activity of the ligand and Pd(II) complex was investigated against T98G human GBM cell line, while healthy fibroblast cells (L-929) was used for biocompatibility. The cytotoxici ties of Ligand and Pd(II) complex, alone or in combination with EP, were determined by MTT test. Results: Pd(II) complex showed good anti-cancer activity in T98G cells (IC50= 394.67), while it didn't show cytotoxic activity in L-929 fibroblast cells (IC50= 1449.28). The ligand showed cytotoxic activity at IC50= 1126.79 and IC50= 1283.36 levels in T98G and L-929 cell lines, respectively.
