Özet: Hepatit B virüs DNA 'sını belirlemekte kullanılan iki yöntem; nested PCR ve Abbott'un sıvı-hibridizasyon yöntemleri karşılaştırıldı. HBsAg'i rastgele pozitif bulunmuş 88 hastayı (22'si HBeAg pozitif, 66'sı HBeAg negatif) her iki yöntem ile çalışıldı. HBV-DNA; HBeAg pozitif hastaların %90'nında PCR ile, %54'ünde Abbott'un sıvı-hibridizasyon yöntemi ile HBeAg negatif hastaların %60'ında PCR ile %15'inde Abbott sıvı-hibridizasyon yöntemi ile pozitif olarak bulundu. HBV-DNA düzeyi Abbot ile 1-20 pg/mL arasında değerlendirilen hastaların sadece %38'inde, HBV-DNA PCR ile pozitif olarak saptanabildi. HBV-DNA belirlemesinde kullanılan Abbott sıvı-hibridizasyon yönteminin 1-20 pg/mL düzeyleri için spesifitesinin düşük ve standardizasyon çalışmalarına gereksinimi olduğu kabul edildi.

Abstract

Summary: We compared two assays detecting hepatitis B virus DNA (HBV); an in-house nested polymerase chain reaction (PCR) and a liquid hybridization assay of Abbott. We studied 88 HBsAg+ve patients' sera which vere randomly collected (of the 88 patients, 22 were HBeAg+ve and 66 were HBeAg-ve). HB V-DNA was found to be positive in 90% and 54% among HBeAg+ve patients by PCR and by Abbott liquid hybridization assay, respectively. The corresponding figures for HBsAg-ve patients were 60% and 15%. Only 38% of patients in which HBV-DNA levels were between 1-20 pg/mL by the Abbott liquid hybridization assay had detectable HBV-DNA by PCR. The specificity of Abbott tests detecting levels of HBV-DNA are rather low and require further standardization studies for the levels between 1-20 pg/mL.
