Acetoin / butanediol production enhancement by microbial hemoglobin (VHb) in the genetically engineered Enterobacter aerogenes

Özet
Enterobacter aerogenes (Ea) ve rekombinantlarında 2,3-bütandiol ve asetoin oluşumuna bakteriyel hemoglobininin (VHb) etkisi test edilmiştir. Ea'ya plazmid vektörü pUC8 eklenerek EapUC8 suşu ve vgb ilavesiylede rekombinant pUC8:16 suşu genetik mühendisliğiyle yoluyla elde edildi. Suşların iki farklı büyüme koşulları fermentasyon aleti kullanılarak incelenmiştir. Üç suşa (Ea, EapUC8, EapUC8:16) ait büyüme özellikleri, pH değişimi ve asetoin/bütandiol üretimi düzenlenen her iki koşul altında ölçülmüştür. Her iki koşulda da VHb'nin asetoin ve 2,3- bütandiol üretimini arttırdığı ve yüksek havalandırma ile daha fazla asetoin ve bütandiol oluşturduğu gözlemlendi. Genel olarak hemoglobin geni hücre büyümesine yardım etmemenin yanısıra oksijen kullanımı ve asit üretiminide önemli oranda etkilemedi.
Abstract
The effect of bacterial hemoglobin (VHb) in the formation of 2,3-butanediol and acetoin was tested in Enterobacter aerogenes (Ea) and its recombinants. Ea was engineered with plasmid vector pUC8 containing the VHb gene (vgb) (recombinant plasmid pUC8:16) to form strain EapUC8:16. Two different growth conditions were examined using a fermenter. Growth proprieties, pH Change, and acetoin / butanediol production of the three strains was measured under both sets of conditions. It was reported that VHb increases the production of acetoin plus 2,3-butanediol under both conditions and that more acetoin and butanediol were produced with higher aeration. The hemoglobin gene did not, in general, aid in cell growth as well as oxygen utilization and acid production.
