Comparison of ATP-MgCl2 and methylprednisolone in experimentally induced spinal cord trauma

Özet

Amaç: Bu çalışamanın amacı, Metilprednizolon sodyum süksinat (MPSS) ile ATP-MgCl2’ün spinal kord travmalarından sonra sekonder hasarın önlenme-sindeki etkinliğinin klinik, elektrofizyolojik ve histopatolojik muayenelerle kar-şılaştırılmasıdır. Gereç ve Yöntem: Çalışmada Wistar-Albino ırkı 30 sağlıklı er-kek rat kullanıldı. Bütün ratlara eğimli yüzey, parmak açma, tarlov testleri uy-gulandı. Anesteziye alınan ratların operasyondan önce Somatosensorik Uyan-dırılmış Potansiyelleri (SEP) ve Motor Uyandırılmış Potansiyelleri (MEP) kay-dedildi. Allen’in ağırlık düşürme yöntemi ile 50g/cm spinal kord hasarı (SKH) oluşturuldu. Ratlar 10’arlı 3 gruba (1. grup FTS, 2. grup MP, 3. grup ATP) ay-rıldı. FTS grubuna fizyolojik tuzlu su, MP grubuna MPSS, ATP grubuna ATP-MgCl2 uygulandı. Operasyondan sonra belirli aralıklarla ratların testleri tek-rarlandı. Operasyondan sonra 72. saatte SEP ve MEP değerleri tekrar kay-dedildi. Travma oluşturulan spinal kord segmenti Hematoksilen - Eozin (HxE) boyası ile boyanarak histopatolojik yönden incelendi. Bulgular: Kalitatif SEP skalası ile Tarlov skalası arasında paralellik izlendi. Bazı elektrofizyolojik bul-gular yönünden MP ve ATP grubu kontrol grubundan (FTS) anlamlı düzeyde iyi bulundu. Bununla birlikte, MPSS ve ATP-MgCl2 uygulanan gruplar arasında belirgin bir fark belirlenmedi. Histopatolojik incelemelerde MPSS’ın polimorf hücre reaksiyonuna neden olduğu izlendi. Sonuç: ATP-MgCl2’ün spinal kord hasarında (SKH) MPSS’dan daha iyi gelişme sağladığını söylemek için daha fazla deneysel çalışmaya gereksinim vardır.

Abstract

Aim: The aim of this study is to compare the preventive effects of methyl prednisolone sodium succinate (MPSS) and ATP-MgCl2 on secondary injury after spinal cord trauma by clinical, electrophysiological and histopathological examination. Material and Method: Thirty healthy male Wistar-Albino rats were used in the study. All rats were evaluated by inclined plane, toe-spread and tarlov tests. Before the operation, somatosensoric evoked potentials (SEP) and motor evoked potentials (MEP) were recorded from anesthetized rats. The spinal cord was traumatised by Allen’s weight drop method applying 50gr/cm force. The rats were divided into 3 groups of 10 members per group (1st group FTS, 2nd group-MP, 3rd group ATP). The first group received saline, the second group received MPSS, and the third group received ATP-MgCl2. At regular intervals after the operation, these tests were repeated on the rats. At the 72nd hour after operation, their SEP and MEP values were recorded again. The damaged spinal cord segment stained with hematoxylin - eosin was evaluated histopathologically. Results: Qualitative SEP scales and Tarlov scales were found compatible. MP and ATP groups were found significantly better than control group (FTS) according to some electrophysiological results. However, a significant difference could not be determined between MPSS and ATP-MgCl2 applied groups. Histopathological examinations showed the presence of MPSS caused polymorphonuclear cell reaction. Discussion: As a result, more experimental studies are needed to say that ATP-MgCl2 is more effective than MPSS in spinal cord injury (SCI).
