Evaluation of a CHROMagar Salmonella Medium for the Isolation of Salmonella Species

Özet

Amaç: Salmonella enfeksiyonları gıda ile bulaşan enfeksiyonların başında gelmektedir ve tüm dünyada gasteroenterit salgınlarına neden olabilmektedir.Salmonella türleri laktozu fermente edememe, tek karbon kaynağı olarak sitratı, nitrat kaynağı olarak lizini kullanma ve TSI agarda Hidrojen Sülfür (H2S)oluşturma yetenekleri ile tanımlanırlar. Ancak yanlış pozitif sonuçların doğrulanması zaman kaybına ve maliyet artışına neden olmaktadır. Bu çalışmada Salmonella türlerinin izolasyon ve saptanması için geliştirilmiş bir besiyeri olan CHROMagar Salmonella (CHROMagar Microbiology, Fransa) besiyerinin değerlendirilmesi amaçlanmıştır. Gereç ve Yöntem: Bu amaçla Akdeniz Üniversitesi Hastanesi Merkez Laboratuvarında çeşitli klinik örneklerdenizole edilerek stoklanmış 65i Salmonella spp., 10u Pseudomonas aeruginosa, 10u E. coli, 10u Acinetobacter baumannii, 10u Klebsiella pneumoniae,18i Morganella morganii, 11i Citrobacter spp, 5i Providencia spp., 4ü Aeromonas spp., 5i Proteus spp. olmak üzere toplam 148 izolat çalışmaya dahiledilmiştir. Bulgular: CHROMagar Salmonella besiyerinde 65 Salmonella spp.izolatının tümü leylak rengi koloniler oluşturmuştur. E. coli ve Klebsiella pneumoniae suşları mavi renkli; Providencia türleri renksiz ve mavi renkli; Morganella morganii suşları renksiz ve açık leylak rengi; Pseudomonas aeruginosa suşları renksiz koloniler oluşturmuştur. Acinetobacter baumannii suşları veAeromonas spp. suşlarının da leylak rengi koloniler oluşturduğu gözlemlenmiştir. Tartışma: CHROMagar besiyeri ile %100 duyarlılıkta Salmonella türleri saptanabilmekte, ancak biyokimyasal veya serolojik doğrulamaya gereksinim duyulmaktadır.

Abstract

Aim: Salmonella infections are the leading cause of food-borne infectionsand can cause gastroenteritis outbreaks worldwide. Salmonella species isdefined as inability to lactose fermentation, using citrate as a carbon source,using lysine as nitrate source and forming Hydrogen sulfide (H2S) in TSI agar.However, confirmation of false positive results is time consuming and leadto increased costs. The aim of this study is to evaluate the performance ofCHROMagar Salmonella (CHROMagar Microbiology, France) which is devel- oped for isolation and detection of Salmonella species. Material and Method:For this purpose, among a total of 148 isolates which were isolated fromvarious clinical specimens and stocked at the Central Laboratory of AkdenizUniversity Hospital, 65 were Salmonella spp., 10 were Pseudomonas aeru- ginosa, 10 were E. coli, 10 were Acinetobacter baumannii, 10 were Klebsi- ella pneumoniae, 18 were Morganella morganii, 11 were Citrobacter spp.,5 were Providencia spp., 4 were Aeromonas spp., 5 were Proteus spp. wereincluded in this study. All of the 65 Salmonella spp. isolates apperared withmauve colonies at the CHROMagar Salmonella. Results: E. coli and Klebsi- ella pnemoniae species were seen as blue, Providencia species were seen aspale-blue; Morganella morganii species were seen as pale-pink, mauve; andPseudomonas aeruginosa species were seen as pale. Acinebacter baumanniiand Aeromonas spp. species were also seen as mauve colonies. Dicussion:CHROMagar Salmonella medium can detect Salmonella species with %100sensitivity, however there is a need to biochemical or serological confirmation.
