Measuring nuclear DNA-content of oral smears using a seescan TV image analysis system (TVIAS)

Özet

Sitofotometre sitolojik oral yayma preparatla-rında DNA-içeriğinin karakterize edilmesinde kullanılmaktadır. Bu çalışmanın amacı üç fark¬lı makinayla ölçülen seçilmiş normal ve anor¬mal yayma preparatlarının DNA-içeriğini kar-şılaştırmakdır. Yirmibeş adet oral yayma pre-paratı a) Vicker's M85 mikrodensitometre b) IBAS görüntü analiz sistemi c) Seescan görün¬tü analiz sistemi kullanılarak ölçüldü. Onbir normal, 5 adet non-displastik ve 5 adet histo-patolojik olarak displastik veya malignant oldu¬ğu onaylanmış yayma preparatı değerlendiril¬di. M85 ve IBAS, M85 ve Seescan sistemleri arasında % 92 ve % 88 uyum, IBAS ve Sees¬can sistemleri arasında % 80 uyum saptandı. Sonuç olarak, Seescan'in M85'ın yerini alan güvenli bir sistem olduğuna karar verildi.

Abstract

Cytophotometry has been widely used to cha¬racterise the DNA-content of cytological sme¬ars. The aim of this study was to compare the DNA content of a selection of normal and ab¬normal smears when measured three different machines. Twenty five oral smears were me¬asured using a) a Vicker's M85 microdensito-meter, b) an IBAS image analysis system, c) a Seescan Image analysis system. Eleven nor¬mal, five non-dysplastic and nine histologically confirmed dysplastic or malignant smears we¬re assessed. There was 92% and 88% agre¬ement between the M85 and the IBAS and Seescan systems, with 80% agreement betwe¬en the IBAS and Seescan systems. It is conc¬luded that the Seescan system provides a reli¬able replacement for M85.
