Özet

Kök kanal tedavisinin başarısını etkileyen en önemli faktörlerden biri de kök kanal dolgu maddeleridir. Canlı dokularla temas halinde olan maddelerin biyolojik yönden uyumlu olmaları gerekir. Kanal dolgu maddeleri canlı dokularla temas halinde olabilir. Bu nedenle biyolojik özelliklerinin araştırılması gerekmektedir. Bu amaçla uygulanan en önemli tekniklerden biri de hücre kültürleri üzerinde yapılan sitotoksik etki testleridir. Bu çalışmada, cam iyonomer esaslı yeni bir madde olan Endion ile Ca(OH)2 esaslı Biocalex ve ZOE içeren Endomethasone'un sitotoksik etkilerinin hücre kültürlerinde karşılaştırmalı olarak incelenmesi amaçlanmıştır. Maddelerin sitotoksitelerinin değerlendirilmesinde VERO (Afrika Yeşil Maymun Böbrek Hücresi) hücre kültürü kullanıldı. Toksisite değerlendirilmesi sırasında, hücrelerdeki morfoloji değişiklikleri, yuvarlaklaşma ve bir araya toplanarak gruplaşmaları göz önüne alındı. Kullanılan kök kanal dolgu maddelerinin sitotoksik etkileri incelendiğinde Endion'un en toksik madde olduğu ve sırasıyla bunu Endomethasone ve Biocalex'in izlediği gözlendi.

Abstract

One of the most important factor that effects the success of root canal therapy is the root canal sealer. The material which contact with the vital tissue must be biocompatible. Root canal materials can be in contact with the vital tissue. Therefore, searching the biocompatibility features is necessary. For this purpose, one of the most important technique is the cytotoxicity test on the cell cultures. The purpose of this study is comparatively evaluating the cytotoxic effects of glass ionomer based Endion, calcium hydroxide (Ca(OH)2) based Biocalex, and zinc oxide eugenol (ZOE) based Endomethasone in the cell cultures. VERO (African Green Monkey Kidney Cells) cells were used for the cytotoxicity evaluation of the materials. The criteria for the evaluation of the cytotoxicity were the morphological changes of the cells, and their collocation. The result showed that Endion was the most toxic one, followed by Endomethasone and Biocalex respectively.
