Özet
Bu çalışmanın amacı oldukça nadir görülen A. actinomycetemcomitans (Actinobacillus actinomycetemcomitans) serotip d ve e için özel PCR (polimeraz zincir reaksiyonu)-bazlı serotiplendirme yöntemini ortaya koymaktır. Çalışmada bütün serotipleri ve 19 AP-PCR (rastgele primerize edilmiş-PCR) genotipi içeren toplam 148 A. actinomycetemcomitans izolasyon serotip d-özel AP-PCR amplikon seçimi için analize edildi. Amplikon tespit edildikten sonra DNA ayrıştırıldı. Klonal fragmant dizilimi elde edildi ve 988-bp iç dizilim gösteren Aad-2 ve Aad4 primer çifti serotip d'nin spesifik tespitine olanak sağladı. Ayrıca 1042-bp özel iç dizim gösteren Aad-3 ve Aad-5 primer çifti ise serotip d ve/veya e'nin saptanmasına imkan verdi. 988-bp'lik fragmant bütün test A. actinomycetemcomitans izolasyonlarından amplifiye edildi. Benzer olarak Aad-3 ve Aad-5 primer çifti sadece serotip d ve e izolasyonlarını tespit etti. Bu sunulan yeni PCR tetkiki A. actinomycetemcomitans'ın farklı laboratuvarlarda standart şekilde serotiplendirilmesine olanak sağlamış ve serotip d ve e grubuna ait izolasyonların tiplendirilmesinde umut verici bir araç olduğu ortaya konmuştur.
Abstract
The aim of the study was to report a novel PCR-based serotyping assay specific for the rare Actinobacillus actinomycetemcomitans serotypes d and e. A total of 148 A. actinomycetemcomitans isolates of all known serotypes and 19 arbitrarily primed polymerase chain reaction (AP-PCR) genotypes were analyzed to select a serotype d-specific AP-PCR amp/icon. An amplicon was identified and DNA was purified. The cloned fragment was sequenced and a primer pair Aad-2 and Aad-4 flanking a 988-bp internal sequence provided specific determination of the serotype d. Another primer pair Aad-3 and Aad-5 flanking a 1042-bp specific internal sequence allowed determination of both serotype d and serotype e. The 988-bp fragment could be amplified from all tested A. actinomycetem-comitans serotype d isolates Similarly, the primer pair Aad-3 and Aad-5 simultaneously detected only all tested serotype d and serotype e isolates. The described novel PCR assay is amenable for standardization for serotype determination of A. actinomycetemcomitans in different laboratories and presently provides a promising tool for typing isolates belonging to serotype d and serotype e groups.
