Özet

Bu araştırmanın amacı, periodontal defektlerin tedavisinde yönlendirilmiş doku rejenerasyonu (YDR) tekniği ile birlikte kullanılabilecek kollagen esaslı membran materyallerinin rezorpsiyon özellikleri ve hücresel cevaplarını bir sıçan modelinde transmisyon elekron mikroskobunda (TEM) incelemektir. Çalışmada 20 adet Wistar sıçanı kullanılmıştır. Ticari olarak elde edilebilen kollagen esaslı iki membran; insan dura mater ve tip l bovin kollagen membran 0.5x0.5cm boyutlarında sıçanların sırtına subkutan olarak implante edilmiştir. Genel anestezi altında 40 implant materyali (20 dura mater ve 20 tip l bovin kollagen) yerleştirilmiştir. Deney hayvanları 14. günde dekapite edilerek ilgili bölgelerden blok biyopsiler alınmış ve elde edilen örnekler membran çevresindeki doku kompozisyonu, fagositik hücrelerin yapısal özellikleri ve membranla-rm rezorbsiyon karakteristikleri yönünden TEM de incelenmiştir. TEM de yapılan incelemeler tip l bovin kollagen membranın dura mater membrana göre daha hızlı rezorbe olduğu şeklinde yorumlanmış ve 14. günde her iki grupta da rezorpsiyon işleminin devam ettiği gözlenmiştir. Ayrıca membran materyallerine karşı istenilmeyen bir doku cevabı tespit edilmemiştir.

Abstract

The aim of the present study was to investigate the resorption characteristics of collagen based membrane materials that might be used for guided tissue regeneration (GTR) procedures in the treatment of periodontal defects, using transmisson electron microscopy (TEM) in a rat model. In the study 20 Wistar rats were used. Two commercially available resorbable collagen based membrane materials human dura mater and type I bovine collagen were implanted subcutaneously on the dorsal surface of rats with the dimension of 0.5x0.5cm. 40 implant materials (20 dura mater, 20 type I bovine collagen) were placed under general anaesthsia. Experimental animals were sacrificed 14 days post-implantation and tissue specimens were evaluated for the composition of the surrounding tissue, the structural patterns of phagocytic cells and the resorption characteristics of the collagen membranes using TEM. TEM examinations were interpreted as the type I bovine collagen membranes were resorbed faster than the dura mater membranes and resorption process was continuing in both groups at 14 days. In addition no adverse issue reactions were determined towards to these membrane materials.
