Neonatal ishalli buzağılarda rotavirus, coronavirus, E. coli K99 ve Cryptosporidium parvum'un hızlı test kitleri ile teşhisi ve enteropatojen ile maternal immünite ilişkisi

Özet
Bu çalışma, neonatal dönemdeki ishalli buzağılarda, ishalin en önemli nedenlerinden olan rotavirus, coronavirus, E.coli F5 (K99) ve Cryptosporidium’un hızlı test kitleriyle tespit edilmesi ve bu enteropatojenler ile serum IgA ve IgG düzeyleri arasındaki ilişkiyi belirlemek amacıyla yapıldı. Çalışmanın hayvan materyalini akut ishalli, 1-28 günlük yaşta 30 buzağı oluşturdu. Gaita örneklerinde rotavirus, coronavirus, Cryptosporidium ve E. coli F5 tespiti ticari in vitro Rapid Diagnostic Test ile yapıldı. Serum IgA ve IgG konsantrasyonları ticari Single Radial Immunodiffusion test kiti ile belirlendi. Bu çalışmada ishalli buzağıların 9’unda (%30) rotavirus, 4’ünde (%13) coronavirus, 5’inde (%17) E. coli F5, 2’sinde (%6) miksenfeksiyon, 10’unda (%33) diğer nedenlere bağlı ishal saptandı. Bu çalışmadaki ishalli buzağıların hiçbirinde Cryptosporidium tespit edilmedi. Serum IgA düzeyleri genellikle kısmi normal veya normalden aşağı, serum IgG düzeyleri genellikle normalden aşağı saptandı. Buzağılarda görülen ishal olaylarında, çabuk sonuç veren test tekniklerinin pratik teşhiste kullanılması yararlı olacaktır. Çünkü bu gibi enfeksiyonların çabuk teşhisleri, ölümleri azaltacak, tedavi şansını artıracak ve böylece hayvan yetiştiricilerini, buzağı ölümlerinden dolayı oluşabilecek, büyük ekonomik kayıplardan kurtaracaktır.
Abstract
In this study, rotavirus, coronavirus, E. coli F5 (K99) and Cryptosporidium which are among the most important causes of diarrhoea in diarrheic calves in the neonatal period was detected with rapid test kits and the relationship between these enteropathogens and serum IgA and IgG levels was determined. Animal material of the study was composed of 30 calves with acute diarrhoea ages of which were ranging between 1 and 28 day. Detection of rotavirus, coronavirus, Cryptosporidium and E. coli F5 in stool samples was done with commercial in-vitro Rapid Diagnostic Test. Determination of serum IgA and IgG levels was determined by commercial Single Radial Immunodiffusion test kit. In this study, rotavirus in 9 (30%), coronavirus in 4 (13%), and E. coli F5 in 5 (17%), mix-infection in 2 (6%), diarrhoea due to other causes in 10 (33%) of diarrheic calves were identified. Cryptosporidium was detected in any of diarrheic calves used in this study. It was found that serum IgA levels were relatively normal or lower. It was determined that serum IgG levels were lower. Use of rapid test kits for practical diagnosis of diarrhea in calves is usefull. Because rapid diagnosis of such infections will reduce the mortality, increase the chances of treatment, and thus animal breeders will prevent large economic losses that may occur due to calf deaths.
