Özet

Araştırmamızda Aydın ili ve yöresinde bulunan çiftliklerdeki mastitisli sığırlardan alınan süt örneklerinde Listeria monocytogenes varlığının fenotipik ve genotipik yöntemlerle araştırılması amaçlanmıştır. Araştırma materyalini Aydın ili ve çevresinde yer alan sığır işletmelerinde bulunan ve mastitis gözlenen 200 sığır oluşturmuştur. Sığırlardan alınan toplam 200 mastitisli süt örneği soğuk zincir altında laboratuara getirilmiştir. Süt örneklerinden fenotipik yöntemlerle L. monocytogenes identifikasyonları yapılmıştır. L. monocytogenes olarak identifiye edilen izolatlar genotipik olarak PCR ile doğrulanmıştır. L. monocytogenes spesifik primerler kullanılarak gerçekleştirilen PCR sonrasında incelenen toplam 11 adet L. monocytogenes izolatının hepsi (%100) inlB geni açısından pozitif olarak saptanmıştır. Sonuç olarak mastitisli sığırlardan alınan süt örneklerinden fenotipik ve genotipik yöntemlerle %5.5 oranında L. monocytogenes identifiye edilmiş ve Aydın ili süt sığırcılığı işletmelerinde L. monocytogenes varlığı ortaya konulmuştur.

Abstract

In our research, it was aimed to investigate the L. monocytogenes from milk samples taken from cattle with mastitis in Aydın province and its presence by phenotypic and genotypic methods. The research material consisted of 200 cattle that suffered from mastitis in cattle farms in and around Aydın province. A total of 200 milk samples with mastitis taken from cattle were brought to the laboratory under cold chain. Identification of L. monocytogenes was carried out by phenotypic methods from the milk samples. The isolates identified as L. monocytogenes were genotypically confirmed by PCR. A total of 11 isolates of L. monocytogenes (100%) confirmed by PCR using L. monocytogenes specific primers and were identified as positive for the inlB gene. As a result, L. monocytogenes were identified by phenotypic and genotypic methods in 5.5% of milk samples taken from cattle with mastitis and L. monocytogenes was identified in Aydın dairy milk cattle farms.
