Doksorubisin’in Sıçan Böbrek Dokularında Yaptığı Hasar Üzerine Vitamin D’nin Etkilerinin ve Asprosin ile ĠliĢkisinin Ġncelenmesi

Özet

Amaç: Bu çalıĢmada doksorubisin kaynaklı böbrek hasarında Vitamin D'nin iyileĢtirici etkileri ve asprosin ile iliĢkisinin araĢtırılması amaçlandı. Gereç ve Yöntem: Erkek Wistar Albino sıçanlar her grupta 6 hayvan olacak Ģekilde 4 gruba ayrıldı. Kontrol grubuna herhangi bir iĢlem uygulanmadı. Vitamin D grubundaki sıçanlara oral olarak 200 IU/gün D Vitamini verildi. Doksorubisin grubundaki sıçanlara deneyin ilk gününde intraperitoneal (i.p.) olarak tek doz doksorubisin 10 mg/kg oranında uygulandı. Doksorubisin+D vitamini grubundaki sıçanlara tek doz doksorubisin i.p. verildi. Deneyin ilk gününde 10 mg/kg ve 14 günlük deney periyodu boyunca her gün oral olarak 200 IU/gün D vitamini verildi. ÇalıĢma sonunda tüm gruplardaki sıçanlar dekapite edilerek doku ve kan örnekleri alındı. Toplam oksidan seviyesi, kan örneklerinden ELISA yöntemi ile ölçüldü. Böbrek dokuları TUNEL ve immünhistokimyasal boyama ile incelendi. Bulgular: Bu çalıĢmada doksorubisinin toplam oksidan düzeylerini ve TUNEL pozitifliğini artırdığı, asprosin düzeylerini düĢürdüğü, tedavi olarak verilen D vitamininin toplam oksidan düzeyini ve TUNEL pozitifliğini azalttığı, asprosin düzeyini değiĢtirmediği belirlendi. Sonuç: Doksorubisin’in asprosini azaltarak doku hasar mekanizmalarında etkili olabileceği ve tedavide D vitamininin iyileĢtirici etkisinin olduğu ancak bu iyileĢtirici etkinin asprosinden bağımsız bir mekanizma ile sağlandığı gösterilmiĢtir

Abstract

Objective: In this study, it was aimed to investigate effects of vitamin D and its relationship with asprosin in rats with doxorubicin-induced kidney damage. Materials and Methods: Male Wistar Albino rats were divided into 4 groups with 6 animals in each group. No procedure was applied to the control group. Rats in the vitamin D group were given 200 IU/day vitamin D orally. A single dose of doxorubicin 10 mg/kg was administered intraperitoneally (i.p.) to the rats in the doxorubicin group on the first day of the experiment. A single dose of doxorubicin i.p. was administered to rats in the doxorubicin + vitamin D group. Vitamin D was given as 10 mg/kg on the first day of the experiment and 200 IU/day orally every day during the 14-day experimental period. At the end of the study, rats in all groups were decapitated and tissue and blood samples were taken. Total oxidant levels was measured from blood samples by Enzyme- Linked ImmunoSorbent Assay (ELISA) method. Kidney tissues were examined by TUNEL and immunohistochemical staining. Results: It was determined that doxorubicin casued increase in total oxidant level and TUNEL positivity, and decrease in asprosin levels, vitamin D given as treatment decreased total oxidant level and TUNEL positivity, and did not change asprosin level. Conclusion: It has been shown that doxorubicin may be effective in tissue damage mechanisms by reducing asprosin, and that vitamin D has a curative effect in treatment, but this curative effect is provided by a mechanism independent of asprosin
