Özet

Paratuberküloz (Johne hastalığı), raminantlann Mycobacterium avium subsp. paratuberculosis ile infeksiyonu neticesinde oluşan kronik, bulaşıcı ve ekonomik olarak önemli bir hastalığıdır. İki koyun ve bir keçiden alman bağırsak ve mezenterik lenf yumrularında paratüberkülozun etkeninin DNA'sini tespit etmek amacıyla sadece bu etkene spesifik bir sekans olan IS9OO'e dayalı polimeraz zincir reaksiyonu (PCR) kullanıldı. PCR ürünlerinin agaroz jel elektroforez ile analizi üç hayvanın da paratuberküloz hastalığına maruz kaldığım ortaya koydu. Bu çalışma, Türkiye'de küçük ruminantların gastrointestinal bölgesinde ve bu bölgedeki lenf yumrularında M. avium subsp. paratuberculosis'm DNA'sının PCR ile tespit edildiğini bildiren ilk çalışmadır.

Abstract

Paratuberculosis (Johne's disease) is a chronic contagious and economically important disease of ruminants associated with Mycobacterium avium subsp. paratuberculosis infection. A polymerase chain reaction (PCR) based on IS900, an insertion sequence that is specific to the causative agent of paratuberculosis, was employed to detect DNA of this organism in the intestines and mesenteric lymph nodes obtained from two sheep and a goat. Agarose gel electrophoresis analysis of the PCR products showed that all three animals had paratuberculosis. This is the first study that reports the detection of M. avium subsp. paratuberculosis DNA by PCR in the gastrointestinal tract and regional lymph nodes of small ruminants in Turkey.
