Investigation of nectin-2 expression in the Rat thymus during organogenesis in the embryo

Özet

Amaç: Timus, T lenfositlerin gelişimi ve olgunlaşması için gerekli mikroçevreyi sağlayan ve epitelyal mezenşimal etkileşimin yoğun olduğu bir primer lenfoid organdır. Nektinler Ca+2-bağımsız immunoglobulin-benzeri hücre adezyon molekülüdür. Bu çalışmanın amacı, gelişimin farklı dönemlerindeki sıçan fetus timusunda nektin-2 ekspresyonunun araştırılmasıdır. Gereç ve Yöntem: Çalışmada 24 erişkin wistar-albino cinsi dişi sıçan kullanıldı. Gebe sıçanlar 14, 16, 18 ve 20. günlerinde ketamin anestezi altında dekapite edildi ve fetusları çıkarıldı. Fetuslar parafin bloklara gömüldü. Genel histolojik yapıyı görmek amacıyla alınan kesitler hematoksilen-eozin ile boyandı. Nektin-2, fetal timusda streptavidin-biotin peroksidaz tekniği kullanılarak immunohistokimyasal olarak belirlendi. Bulgular: Timus taslağı gebeliğin 14. ve 16. gününde bağ dokusu kapsülü ile çevrelenmişti. Gebeliğin 18. gününde bağ dokusu kapsülünün oluşturduğu septa dokuyu tam olmayan lobüllere ayırdı. Lobulasyon gebeliğin 20. gününde daha be-lirgindi. Nektin-2 immunreaktivitesi gebeliğin 14. ve 20. gününde orta yoğunlukta, gebeliğin 16. ve 18. gününde zayıf reaksiyon gösterdi. Sonuç: Timopoezis sırasında hücre-hücre etkileşimlerindenektin-2’nin katkısı olduğu düşünülmektedir.

Abstract

Objective: The thymus is a primary lymphoid organ which provides the essential microenvironment for T lymphocyte development and matures through epithelial-mesenchymal interactions. Nectins are Ca2+-independent Ig-like cell adhesion molecules. The purpose of this study was to investigate nectin-2 expression in the thymus at different stages of development in the rat foetus. Material and Method: In this study, 24 pregnant adult female Wistar-albino rats were used. The rats were decapitated under ketamine anaesthesia and their foetuses were removed at 14, 16, 18, and 20 days of gestation (GD). Foetuses were embedded in paraffin blocks. In order to determine the general histological structure, sections were stained with hematoxylin- eosin. Nectin-2 was detected in the foetal thymus by immuno- histochemistry using the avidin-biotin-peroxidase technique.
