Özet

Serbest oksijen radikalleri, membran yapısındaki lipid ve proteinlerin oksidasyonu yolu ile doku hasarının patolojisinde rol oynar. Bu çalışmanın amacı hiper- ve hipotiroidili hastalarda lipid peroksidasyon, protein oksidasyon ve antioksidan düzeyleri araştırmaktır. Kadınlardan oluşan 22 hipertiroid, 15 hipotiroid ve 15 sağlıklı kişi çalışma kapsamına alındı. Kontrol grubu ile karşılaştırıldığında hipertiroidili ve hipotiroidili hastaların eritrosit ve plazma tiyobarbiturik asitle reaksiyonlaşan maddeler (TBARS) düzeyi (lipid peroksidasyonunun bir göstergesi) ve eritrosit protein karbonil içeriği (protein oksidasyonunun bir göstergesi) anlamlı derecede yüksek , antioksidan olan eritrosit glutatyon (GSH) düzeyi ve glutatyon peroksidaz aktivitesi (GSH-Px) ve plazma total tiyol düzeyi anlamlı derecede düşük bulundu. Hipotiroidili hastalar hipertiroidili hastalardan anlamlı derece daha yüksek eritrosit TBARS ve plazma kolesterol ve daha düşük GSH düzeyine sahipti. Bu bulgular oksidatif stresin tiroid hormon bozukluklarının yol açtığı hücresel hasarlarda rol aldığını göstermektedir.

Abstract

Oxygen free radicals play a role in the pathogenesis of tissue damage via the oxidation of both membrane lipids and proteins. The purpose of this study was to examine lipid peroxidation, protein oxidation and antioxidant states in hyper-and hypothyroid patients. We examined 22 hyperthyroid patients, 15 hypothyroid patients and 15 healthy controls. All subjects were women. Both in the hyperthyroid and hypothyroid patients plasma and erythrocyte tiobarbituric acid ractive substance (marker of lipid peroxidation) and erythrocyte protein carbonyl content (marker of protein oxidation) were significantly high and erythrocyte glutathione (GSH) levels and glutathione peroxidase (GSH-Px) activity, and plasma total thiols levels (antioxidants) were low as compared to the control group Hypothyroid patients have significantly higher erythrocyte TBARS and plasma total cholesterol levels and significantly lower GSH content than the hiperthyroid patients. These findings suggest that free radicals was involved in tissue damage due to the thyroid hormone disorder.
