The availability of 8-ohdg, an oxidative DNA damage marker, in the diagnosis and treatment of autoimmune thyroiditis

Bu çalışmada, ELISA yöntemi kullanılarak, Hashimoto tiroiditi (n:13, yaş: 48.0 11.8 yr) ve Graves hastalığı (n:7, yaş: 39.5 13.3 yr) olan hastalarda bir DNA hasar biyomarkörü olan 8-hydroxydeoxyguanosine (8-OHdG) miktarı sağlıklı bireylerden oluşan kontrol grubundan elde edilen değerler ile karşılaştırılmıştır (n:15, yaş: 39.1 7.3 yr). Hashimoto tiroiditi (0.404 0.25 ng/ml) ve Graves hastalığı (0.375 0.39 ng/ml) olan hastaların serumlarındaki 8-OHdG seviyesi her ne kadar kontrol grubundan (0.277 0.09 ng/ml) elde edilen değerlerden yüksek olsa da, bu fark istatiksel olarak anlamlı bulunmamıştır. Otoimmün tiroiditi tedavisinin oksidatif DNA hasarı üzerindeki etkisini belirlemek için toplam 10 hastanın serumundaki 8-OHdG seviyesinde tedavi öncesi ve tedavi sonrası arasındaki fark belirlenmiştir. Hashimoto tiroiditi ve Graves hastalığı olan hastalardaki 8-OHdG seviyeleri hipotiroid koşullarda tiroid hormon tedavisi ya da hipertiroid koşullarda propyciltiouracil tedavisi neticesinde önemli ölçülerde azalmış (tedavi öncesi, 0.316 0.07 ng/ml; tedavi sonrası, 0.229 0.07 ng/ml) ve 8-OHdG düzeyleri arasında anlamlı bir fark gözlemlenmiştir (p= 0,016). Elde edilen bu sonuçlar serumdaki 8-OHdG miktarının otoimmün tiroiditi hastalığı olan hastalarda oksidatif DNA hasar biyomarkörü olarak kullanılabileceğini önermektedir. Sonuç olarak, tiroid oksidatif stres üzerinde güçlü bir etkiye sahip gibidir ve otoimmün tiroiditi hastalığının tedavisi sistemik oksidatif stresi azaltabilir.
Abstract
In this work, by using ELISA method, the amount of 8-hydroxydeoxyguanosine (8-OHdG), a DNA damage marker, in serum of patients suffering from Hashimoto's thyroiditis (n:13, age: 48.0 11.8 yr) and Graves' disease (n:7, age: 39.5 13.3 yr) was compared with that of control group containing healthy peoples (n:15, age: 39.1 7.3 yr). Although the level of 8- OHdG in serum of patients with Hashimoto's thyroiditis (0.404 0.25 ng/ml) and Graves' disease (0.375 0.39 ng/ml) were found to be higher than that of control group (0.277 0.09 ng/ml), this difference was not statistically significant (p=0.36). To determine the effect of treatment of autoimmune thyroiditis on the oxidative DNA damage, 8-OHdG levels in serum of ten patients were determined before and after treatment. 8-OHdG levels in patients with Hashimoto's thyroiditis and Graves' disease were significantly decreased by treatment with thyroid hormone in hypothyroid condition or propyciltiouracil in hyperthyroid condition (0.316 0.07 ng/ml, 0.229 0.07 ng/ml, before and after treatment respectively, p= 0.016). These results suggest that the amount of 8-OHdG in serum could be a useful biomarker of oxidative DNA damage in patients suffering from autoimmune thyroid disease. It is our conclusion that thyroid seems to have a potent influence on oxidative stress and treatment of autoimmune thyroid disease may reduce systemic oxidative stress.
