Özet

Bu çalışmada, Artemisia annua methanol ekstresinin Allium cepa kök meristem hücreleri üzerindeki sitotoksikve genotoksik etkileri incelenmiştir. Soğanlar ekstrenin 450, 900, 1350 ve 1800 mg/mL konsantrasyonlarına3, 6 ve 12 saat süre ile maruz bırakılmıştır. Saf su bir negatif kontrol olarak, etil metansülfonat ise bir pozitifkontrol olarak kullanılmıştır. Test konsantrasyonları alternatif tıpta kullanılması önerilen dozlar ve onlarınkatlarına göre belirlenmiştir. Kullanılan ekstre konsantrasyonlarıyla soğan köklerinde, kök uzamasınınazalması ve renk değişimi gibi morfolojik değişiklikler takip edilmiştir. Mitotik indeks kontrol gruplarındave test gruplarında belirlenmiştir. Genellikle, mitotik indeks artan ekstre konsantrasyonuyla azalmıştır.Mitotik anormallikler; düzensiz profaz, c-mitoz, başıboş kromozom, yapışıklık, ileri giden veya geri kalankromozom ve anafazda fragment oluşumu olarak kaydedilmiştir. Bu makalede, Artemisia annua ekstresiuygulamasıyla kromozomal anormalliklerin oluşumu, hücre ölümü ve hücre zarındaki bozukluklar ilk kezgösterilmiştir. Böylece, farklı testlerle A. annua ekstresinin sitotoksik ve genotoksik potansiyeli A. cepa kökhücrelerinin kullanılmasıyla değerlendirilmiştir ve toksik olmadığı belirlenen dozun kullanımı önerilmiştir.

Abstract

In this study, the cytotoxic and genotoxic effects of Artemisia annua methanol extract on the root meristemcells of Allium cepa was investigated. The onion bulbs were exposed to 450, 900, 1350, and 1800 mg/mLconcentrations of the extract for 3, 6, and 12 h. Distilled water was used as a negative control and ethylmethanesulfonate was used as a positive control. The test concentrations were determined according to thedoses which are suggested for utilization in alternative medicine and it's multiple. The occurringmorphological changes such as reduction of root elongation and discoloration in the onion roots using theextract concentrations are as follows. The mitotic index was determined in both controls and test groups.Generally, the mitotic index decreased when increasing the extract concentration. The mitoticabnormalities were recorded as disturbed prophase, c-mitosis, vagrant chromosome, stickiness, going aheadchromosome, laggards, chromatid bridges, and fragment formation in the anaphase. In this manuscript, theoccurrence of chromosomal abnormalities, cell death, and impairments in the cell membrane by Artemisiaannua extract has been indicated for the first time. Thus, the cytotoxic and genotoxic potency of A. annuaextract with different tests were assessed by using the root tip cells of A. cepa, and the use of a determinednon-toxic dose was recommended.
