Triticum dicoccoides ile Triticum durum melezlerinin $F_4$ neslinde ve ebeveynlerinde RAPD yöntemiyle genotip relirlenmesi

Özet
Buğday insan beslenmesinde önemli bir bileşendir. Bu nedenle buğdayın kalite ve kantitesini artırmak için birçok ıslah çalışması yapılmaktadır. Bugün genotipik yapıların gözlemini mümkün kılan çeşitli moleküler markörler kullanımdadır. Bu araştırmada, makarnalık buğday (Triticum durum) ve yabani buğday (Triticum dicoccoides) ekonomik beslenme ve yüksek protein içeriği açısından önemleri nedeniyle kullanıldı ve bu türlerin hibritleri RAPD markırları kullanılarak analiz edildi. Yirmi RAPD primerinin ön analiz sonuçlarına dayalı olarak önemli bantlar veren 6 primer (OPA-02, OPA-04, OPA-06, OPB-03, OPB-04, OPB-05) genetik polimorfizmi ve genotipleri belirlemek için seçilip kullanıldı. Elektroforetik bantlar Kluster yöntemine göre (jump) analiz edildi. Türler arasındaki genotip uzaklıklar dendogramlarla belirlendi. Araştırma sonucunda, Triticum dicoccoides’ in diğer kültür ebeveynleri ve melez döllerinden ayrı bir grup oluşturduğu gözlendi. Bu veriler gelecek ıslah çalışmaları için bir temel oluşturmaktadır.
Abstract
Wheat is an important element of the human diet. For this reason, in order to increase the quality and the quatity of the wheat, many breeding studies are breing carried out. Today, the various molecular markers that enables the observation of the genotype structures are in use. In this research, the durum wheat (Triticum durum) and wild wheat Triticum dicoccoides were used because of their economical and dietary importance and high protein content respectively. Their hybrid offsprings were analyzed by using RAPD markers. Six primers providing significant bands (OPA-02, OPA-04, OPA-06, OPB-03, OPB-04, OPB- 05) were selected and used in RAPD analysis to determine the genetic polymorphism and identifying the genotypes, based on preliminary results of 20 RAPD primers. The electrophoretic data were analyzed by cluster method (Jump). Genotype distances among the species were revealed by dendograms. According to the results, it was determined that T. dicoccoides formed an other group different from the cluster parents and hybrid offsprings. This data forms a basis for the future breeding studies.
