Abstract

Diamine oxidase was purified 1865 fold from roots of peanut seedling using a new affinity material of histamine bound to porous glass. The purified enzyme was found as a monomer with a molecular weight of 66 kD a. The Km and Vmax for histamine were found to be 1.1x$10^{-3}$ M and 0.83 $mu$mol/mg.min. respectively. The optimum, temperature and pH were 37°C and 7.0. Furthermore, the effect of various inhibitors such as the copper binding reagent, diethyldithiocarbamate, carboxyl and imidazole group specific reagents on the enzyme activity was also studied.
