Özet

Bu çalışmada idrar yolu enfeksiyonlu (İYE) çocuklarda lökositlerin trombosit aktive edici faktör (PAF) sentez ve salgılama aktivitesi değerlendirilerek akut inflamatuvar yanıtın başlatılması ve ilerlemesinde PAF'ın İYE patogenezindeki rolü araştırılmak istenmiştir. Çalışmaya İYE'lu 33 çocuk (32 kız, 1 erkek, yaş ortalaması 6.5 ± 2.8 yıl) alındı. Yirmi iki sağlıklı çocuktan (6 erkek, 16 kız, yaş ortalaması 5.7 ± 3.1 yıl) kontrol grubu oluşturuldu. İYE'lu çocuklar 38.5°C ve üzerinde ateş yüksekliği, C-reaktif protein ve sedimantasyon yüksekliği ve DMSA sintigrafi bulgularına göre alt ve üst İYE olarak gruplandırıldı. Endojen plazma ve idrar PAF düzeyleri solid faz ekstraksiyon yöntemi ile purifikasyon sonrası RIA ile ölçüldü. Periferik kan ve idrardan elde edilen lökositler invitro uyarılarak intrasellüler ve ekstrasellüler PAF düzeyleri ölçüldü. Abstract

The purpose of this research is to determine the role of platelet activating factor (PAF) in urinary tract infection (UTI); peripheral blood leukocytes and infiltrated ones in the urinary tract were investigated for PAF synthesis and secretion by in vitro stimulation due to UTI. Thirty-three children (1 boy and 32 girls, mean age 6.5 ± 2.8 years) with UTI were included as the UTI test group in this study. Twenty-two children (6 boys and 16 girls, mean age 5.7 ± 3.1 years) with no UTI acted as the controls. RIA determined PAF concentrations in urine and plasma after purification with solid phase extraction method. Peripheral and urine leukocytes were stimulated in vitro and the cell associated and the released PAF in response to stimulation were determined.
