Bazı domates hatlarının domates lekeli solgunluk virüsü (TSWV=Tomato spotted wilt virus)'ne karşı reaksiyonlarının mekanik inokulasyon yöntemi ile belirlenmesi

Özet
Son yıllarda dünyada ve ülkemizde domates yetiştirilen alanlarda sıkça görülen domates lekeli solgunluk virüsü (Tomato Spotted Wilt Virus=TSWV), %42.1 ile %95.5 arasında ürün kayıplarına neden olabilmektedir. Hastalık etmeni virüs ile mücadelede en etkin yöntem genetik dayanıklılıktır. Bu hastalığa karşı dayanıklılık, Solanum peruvianum türünden elde edilmiş dominant tek bir gen olan Sw-5 geni ile idare edilmektedir. Bu çalışmada, ıslah çalışmalarında pratik olarak kullanılabilecek mekanik inokulasyon yöntemi ile hastalığa dayanıklılıkları farklı beş farklı domates genotipi testlenmiştir. Çamköy-Antalya yöresinden elde edilen TSWV izolatı (TSWV- 15) ile yapılan mekanik inokulasyon çalışmaları sonucunda hassas ve dayanıklı bireyler tespit edilmiştir. Testlemelerde dayanıklı materyal olarak, Sw-5 geni ihtiva ettiği bilinen LA 3667 çeşidi kullanılmıştır. Çalışmada kullanılan diğer materyaller Çeşit-1 (dayanıklı ticari çeşit; FORMULA F1), Çeşit -2 (hassas ticari çeşit; BATEM ÖZÇELİK F1), Hat-1(BATEM-114) ve Hat-2 (BATEM-115)’ dir. Virüsün mekanik inokulasyonu sonucunda Çeşit-1 ve LA 3667, TSWV-15 izolatına karşı dayanıklı bulunmuş; Çeşit-2, Hat-1 ve Hat-2’nin ise hassas olduğu tespit edilmiştir. Antalya yöresinden izole edilen virüs izolatına karşı Sw-5 genini bulunduran domates çeşitlerinin dayanıklılık gösterdiği; geliştirilecek dayanıklı çeşitlerde gen kaynağı olarak Sw-5 genini bulunduran çeşitlerin kullanılabileceği anlaşılmıştır.
Abstract
In the tomato raising fields of Turkiye and in all around the world, TSWV - which is seen widely, caused the 42.1% to 95.5% of the product loss, recently. The most effective fight against the disease agent virus is the genetical resistance. The resistance to this disease is managed by the dominant Sw-5 gene, which is obtained from Solanum peruvianum. In this study, five tomato genotypes have different characteristics against virus disease were examined by the mechanical inoculation method-which can be used in breeding programms practically. Sensitive and resistant individuals were determined at the end of the mechanical inoculation studies by using the TSWV isolate (TSWV-15) which was obtained from Çamköy- Antalya region. In the testing period, as a resistant material, LA 3667-which contained Sw-5 gene was used. The other materials those were used in the study were, Çeşit-1 (resistant commercial variety; FORMULA F1), Çeşit-2 (a commercial variety; BATEM ÖZÇELİK F1 ), Hat-1 (BATEM-114 ) and Hat-2 (BATEM-115) (BATEM lines- the reaction against the disease are not determined). As a result of the testing study, Çeşit-1 and LA 3667 were assigned to be resistant to the TSWV-15 isolate. But, Çeşit-2, Hat-1 and Hat-2 were determined to be sensitive. This study is considered to be qualified enough to use for the future improvement programs for the resistance to the disease agent virus.
