Özet

Bu çalışmada, Staphylococcus aureus ile oluşturulan deneysel invivo infeksiyona karşı vasküler politetrafloroeti-len (PTFE) greftlerin direncini arttırmada Fibrin Glue-Rifampisin (FG-R) süspansiyonu uygulamasının etkinliği araştırıldı. Üç ayrı grupta toplam 30 adet Wistar Albino türü erkek sıçan kullanıldı. Grup l'de (kontrol grubu) (n:10), 5x5 mm. boyutta PTFE greft parçaları sıçanların sırt bölgesine, cilt altına yerleştirildi. Grup ll'de (n:10) greft parça¬ları yerleştirilmeden önce FG-R süspansiyonu ile kaplandı. Grup IN'de (n:10) ise konağa bağlı faktörleri ortadan kaldırmak amacıyla hem FG-R ile muamele edilen, hem de muamele edilmeyen greft parçaları aynı sıçanın sırtın¬da iki ayrı bölgede cilt altına yerleştirildi. 1 mi. serum fizyolojik içinde süspanse edilen 1x106 koloni oluşturan bi¬rim (KOB)/ml S. aureus ATCC 25923 susu tüm gruptaki sıçanlara intraperitoneal olarak verildi. Greftler 72 saat sonra çıkarıldı. I. gruptaki tüm greftlerde kültür pozitif bulunurken, II. grupdaki greftlerin tümünde kültür negatifti (p= 0.00001). III. grupta ise, FG-R süspansiyonu ile muamele edilen greftlerin hepsinde kültür negatif ve herhangi bir işleme tabi tutulmayan greftlerin dokuzunda kültür pozitifti (p=0.00006). Sonuç olarak FG-R süspansiyonu uygula¬ması, S. aureus ile oluşturulan deneysel erken greft infeksiyonuna karşı PTFE greftlerin direncini anlamlı dere¬cede arttırdı.

Abstract

The effectivity of Fibrin Glue-Rifampicin (FG-R) suspension application on the resistance of vascular polytetraflu-oroethylene (PTFE) grafts to experimental invivo infection with Staphylocccus aureus ATCC 25923 was investi¬gated in this study. A total of 30 Wistar Albino rats were used in three different groups. In group I (control group, n:10) 5x5 mm. of sterilized PTFE graft pieces were implanted subcutaneously to the back of the rats. In group II (n:10), graft pieces were sealed with FG-R suspension before implantation. In group III (n:10), both the FG-R tre¬ated and untreated graft pieces were implanted subcutaneously in two different places on the back of the rats to eliminate the factors depending on the host. 1x106 colony forming units (CFU)/ml of S. aureus type ATCC 25923 suspended in 1 ml. of saline solution was given intraperitoneally in all groups of rats. Grafts were removed after 72 hours. All of the grafts in group I were culture positive while all were culture negative in group II (p=0.00001). In group III, the grafts treated with FG-R suspension were all culture negative and 9 of the grafts that no procedu¬re carried out were culture positive (p=0.00006). As a result, the application of FG-R suspension was significantly improved the resistance of PTFE grafts to experimental early graft infection with S. aureus.
