Ratlarda geçici abdominal aort oklüzyonu sonrasında pluşan renal hasarın azaltılmasında askorbik asidin rolü

Özet
Amaç: Çalışmanın amacı, distal abdominal aortada akut oklüzyon sonucunda ortaya çıkan geçici iskemi ve reperfüzyonun böbrek dokusu üzerinde yaptığı hasarı ve askorbik asidin bu hasarı önlemedeki etkisini incelemektir.Materyal ve Metod: Çalışmaya alınan 19 adet rat üç gruba ayrıldı. Kontrol grubundaki raflarda (Grup 1), hiçbir girişim yapılmadan batın açıldı ve tekrar kapatıldı, iskemi reperfüzyon (İ/R) grubunda (Grup 2) infrarenal aortaya klemp konularak 3 saat iskemi ve 1 saat reperfüzyon oluşturuldu. Askorbik asit verilen ratlarda da (Grup 3) aynı işlem yapıldı, ancak 5 gün önceden askorbik asit verilmeye başlandı. Ratlardan doku örnekleri alındı ve hemotoksilen eosin boyası ile boyandı. Böbreklerdeki patolojik değişikliklerin derecesini ifade edebilmek için, Evre O ile 4 arasında değişen puanlama sistemi kullanılarak hasar skoru hesaplandı. Sonuçlar: Kontrol grubuna göre, 2. ve 3. gruplarda bulanık şişme, ödem ve konjesyon daha belirgindi. Kontrol grubunun hasar skor puanı 0.6, 2. grubun 2.14, 3. grubun 1.85 olarak hesaplandı. Abstract
Objective: The aim of this study was to investigate renal injury due to ischemia/reperfusion (I/R) after transient acute distal abdominal aortic occlusion and to evaluate the effect of ascorbic acid on these alterations. Material and Method: Nineteen rats were divided into three groups. Rats in the control group (Group 1) were undertaken laparotomy and closed after exploration of abdominal aorta. In the I/R group (Group 2), the infrarenal aorta of the rats were clamped for a period of 3 hours. After the ischemic period, aortic clamping was removed and waited for 1 hour of reperfusion period. The rats in the ascorbic acid group (Group 3) were administered sodium ascorbic acid commencing from 5 days before experimental procedure. The animals were sacrificed and the histological specimens of the lungs were examined. The pathological changes in the lungs were ranged between stage 0+ and 4 correlated to the level of increasing injury.Result: Edema, congestion and hydropic degeneration were more prominent in the specimens of group 2 and 3 as compared to the control group.
