Özet

Yerel mısır genotipleri genetik çeşitliliğin muhafazası için değerli gen kaynaklarıdır. Bu popülasyonların ticari değere sahip özellikler bakımından karakterizasyonu önem taşımaktadır. Günümüzde nişasta özellikle sanayi ürünü olarak değerlendirilmekle birlikte pek çok farklı alanda kullanılmaktadır. Glukoz ve fruktoz ise tatlandırıcı sektöründe yoğun kullanılan ve tatlandırıcı şurup üretimi için vazgeçilmez olan iki bileşendir. Bu araştırmanın amacı daha önce nişasta, glukoz ve fruktoz özellikleri bakımından hiç incelenmemiş yüz elli yerel köy popülasyondaki değişimi tespit etmektir. Varyasyonu belirleyebilmek için mısır tanelerinde bulunan nişasta, glukoz ve fruktoz bileşenlerinin miktar tayini spektorofotometrik yöntemler kullanılarak spektorofotometre ve microplate ile gerçekleştirilmiştir. Laboratuvar analizleri sonucu elde edilen verilerin istatistiksel analizleri augmented deneme desenine uygun olarak analiz edilmiştir. Yerel mısır genotiplerinin sahip olduğu ortalama nişasta oranı %68,31 fruktoz oranı %0,22 ve glukoz oranı %0,63 olarak tespit edilmiştir. Bu genotiplerin ilerleyen yıllarda yapılacak ıslah çalışmalarında gen kaynağı olarak değerlendirilebileceği düşünülmektedir.

Abstract

Local maize genotypes are valuable gene sources for conservation of genetic diversity. The characterization of these populations in terms of commercially valuable characteristics is important. Today, starch is considered as an industrial product, and it is used in many different fields. Glucose and fructose are two components that are used extensively in the sweetener industry and are indispensable for the production of sweetener syrup. The main purpose of this study is to detect the variability in one hundred and fifty landraces originated from different regions of Anatolia that have never been studied in terms of starch, glucose and fructose traits. In order to determine the variation, the concentrations of starch, glucose and fructose components available in the maize kernels were determined by spectrophotometric methods using spectrophotometer and microplate instruments. The statistical analysis of the data obtained as a result of the laboratory analyses was done according to Augmented Experimental Design. Among the landraces, the average starch ratio was 68.31%, the fructose ratio was 0.22% and the glucose ratio was 0.63%. It is thought that these genotypes may constitute good sources of genes that will help breeding studies in the coming years
