Özet
MSUD (maple syrup urine disease) dallı zincirli α -ketoasit dehidrogenaz enzim kompleksinin yapısına giren altı gen ürününden herhangi birisindeki mutasyon sonucu oluşmaktadır. Bu çalışmada, MSUD hastalığı ile ilgili olarak Akdeniz ülkelerinde sık görüldüğü ileri sürülen E1β/80del 11 mutasyonu ile daha önce değişik toplumlarda tanımlanan E1α/887del8 ve G245R mutasyonların 12 MSUD hastasında varlığı araştırıldı. Çalışma sonucunda 12 hastanın üç mutasyonu taşımadığı belirlendi. Çalışma İtalyan araştırmacıların E1β/Ekzon 1'deki 11 baz çiftlik delesyonun Akdeniz ülkelerinde sık görülebileceği öngörüsünü Desteklememektedir. Çalışmamız Türk MSUD hasta genotiplerinin diğer toplumlardan farklı olabileceğini göstermiştir.
Abstract
Maple Syrup Urine Disease (MSUD) is an autosomal recessive metabolic disease with a worldwide distribution of the disease stated to be 1 in 185,000 newborns. We screened three mutations defined in branched chain ketoacid dehydrogenase complex in 12 Turkish patients: E1α/887del7, G245R and E1β/80del11. We did not detect these mutations in our patients. E1β/80del11 was demonstrated in three Japanese patients and in five Italians two of whom had MSUD. It has been suggested that 11 bp deletion in E1p may be relatively prominent in southern European populations. We did not detect this deletion in any of our 12 patients. In conclusion, this study shows that the genotypes of MSUD patients of Turkish origin are different from those of patients of other southern European countries further studies are required to delineate the mutation spectrum in Turkish patients with MSUD.
