Tebuconazole’un in vitro Koşullarda Fusarium culmorum’un Gelişimine ve Konidi Çimlenmesine Etkilerinin Saptanması

Özet
Bu çalışmada, Çanakkale İli buğday alanlarındaki hastalıklı bitki örneklerinden izole edilmiş olan 10 farklı Fusarium culmorum (W.G. Smith) Sacc. izolatına tebuconazole etkili maddeli fungisitin in vitro’daki etkisini araştırmak hedeflenmiştir. Bu amaçla tebuconazole farklı konsantrasyonlarda PDA besin ortamına eklenmiş (0.01, 0.05, 0.1, 0.5, 1, 5, 10, 50, 100, 500 ppm) ve fungusun misel gelişimi, konidi çimlenme yüzdesi, çim tüpü uzaması ve çim tüplerinde görülen değişimler gözlemlenmiştir. Yapılan çalışmaların sonucunda tebuconazole’ün 1 ppm üzerindeki dozlarda miselyal gelişimi yavaşlattığı, 10 ppm üzerindeki dozlarda ise tamamen durdurduğu tespit edilmiştir. Fungisitin konidi çimlenmesine ciddi olumsuz bir etkisi olmadığı, ancak çimlenen konidilerde çimlenme olsa bile doz artışına bağlı olarak morfolojik bozulmalar olduğu tespit edilmiştir. Farklı saat aralıklarında yapılan çim tüpü ölçümlerinde ise tebuconazole uygulaması ile 5 ppm ve üzeri dozlarda çim tüplerinde oluşan bozulmalar ile zaman faktörü ilişkisi belirgin olarak ortaya konmuştur. Yapılan bu çalışma tebuconazole’un konidi çimlenmesini sadece çok yüksek dozlarda engellediği, fakat bunun aksi olarak doz arttıkça koloni gelişimini ve çimlenme tüpünü negatif yönde etkilediği gözlenmiştir. Elde edilen bulgular doğrultusunda iklim koşullarına, buğdayın dayanıklılığına, ilacın uygulanma şekline göre değişmekle beraber farklı dozlarda tebuconazole ile buğday bitkisinde F. culmorum infeksiyonlarına karşı koruma sağlayabileceği ortaya konmuştur.
Abstract
This study was conducted from April to September 2015 in three apple orchards in Ahmetçeli village of Bayramiç district of Çanakkale province. Identification of weed species in apple orchards has been made throughout the gardens. The % coverage in the garden of weed species was investigated in the area 10x4m2 of 1 no orchard, in the area 3x4m2 of 2 and 3 no orchards. Aphid species were recorded in the places designated for the % coverage and 10 shoots in 4 directions of the apple trees of next to these areas. As a result, 55 weed species belonging to 17 families in apple orchards were determined. The most number of weed species was Asteraceae (13 species), Poaceae (12 species) and Fabaceae (6 species) respectively. The species with the highest % coverage area was Medicago polymorpha L. (69%), Lolium perenne L. (62.3%), Equisetum ramosissimum Desf. (58%) and Cyperus rotundus L. (51.3%). Presence of the weed species, 74% were dicotyledonous, except for Erigeron canadensis and Tribulus terrestris L., have ended from June. At the beginning of the season, the dicotyledonous weeds and from June the monocotyledonous weeds became dominant. A total of 8 aphids were identified on weeds and apple trees in the apple orchard. Dysaphis devecta (Walk.), Dysaphis plantaginea (Passerini) and Macrosiphum rosae L. of the aphid species were obtained on the apple trees. Other five species, Hyperomyzus lactuca L., on Sonchus asper (L.) Hill. subsp. glaucescens (Jord.) Ball. and Tragopogon porrifolius L. subsp. longirostris (Sch. Bip.) Greuter; Brachycaudus tragopogonis Kaltenbach, on Tragopogon porrifolius L. subsp. longirostris (Sch. Bip.) Greuter, Brachycaudus helichrysi Kaltenbach, on Erigeron canadensis L. Uroleucon jaceae L. on Centaurea solstitialis L. and Aphis fabae Scopoli on Daucus carota L. were recorded.
