Mezenterik iskemi-reperfüzyon hasarında siklosporin-A, streptokinaz ve ibuprofen'in etkileri

Özet
Çalışmamızda sıçanların mezanter arter köküne 60 dakika süre ile mikroklipslerin konulmasıyla elde edilen iskemi-reperfüzyon hasarı modelinde siklooksijenaz inhibitörü ibuprofen (İBU), immunosupresan ve nötrofil infiltrasyonu engelleyici siklosporin A (CyA) ve trombolitik bir ajan olan streptokinaz'ın (Str) etkilerini araştırdık. İskemi-repeıfüzyon hasarında sakrifiye edilen sıçanlardan hücre yıkımını gösteren kan parametreleri SGOT, SGPT, LDH ve alkalen fosfataz (ÂP), lipid metabolizmasının son ürünü olan Malonildialdehit (MDA) kriter seçilmiştir. Ay¬rıca histopatolojik değerlendirme ile spesifik ve non-spesifik barsak bulguları skorlanmıştır.Biyokimyasal parametrelerde önemli istatistiksel sonuçlar elde edilmemiş, histopatolojik olarak da anlamlı sonuçlar alınamamıştır. Buna karşın MDA değerlerinde serum fiz¬yolojik grubuna göre istatistiksel değerlendirmeler anlam¬lı bulunmuştur.Her üç ajanın birlikte verilmesiyle oldukça anlamlı farklı¬lık saptanmıştır. Bu üç ajanın birlikte kullanılmasının etki¬lerini arttırdığı sonucuna varılmıştır.
Abstract
The Effects of Cyclosporin A, Streptokinase and ibupro¬fen on Mesenteric Ischemia and Reperfusion Injury In this model of ischemia!reperfusion by 60 minute clip¬ping of rat mesenteric arteries we investigated the effects of cyclooxygenase inhibitor (Ibuprofen-IBU), immunosup-ressor and inhibitor of neutrophil activity (cyclosporine A-CyA) and a trombolytic agent (Streptocynase-Str) on in¬jury. The rats were sacrified after two hours of ischemia! reperfusion and the following indices were evaluated. The biochemical indices of cell destruction SGOT, SGPT, LDH and alcalen phosphatase, the end product of lipid metabolism malonyldialdehyde (MDA) and hystopatholo-gical changes were recorded. In analysis of results no significant change was found in biochemical indices and hystopathology. But significant changes in MDA were recorded when compared to saline control group. The most significant change was observed in the group where three agents were used together. And it was concluded that when these three agents were used to¬gether their effects on ischemialrepeifusion injury were potentiated.
