E.coli ile oluşturulan deneysel peritonitte E vitamini ve N-asetil sistein'in etkileri

Bakteriyel, peritonit ve bunun sonucunda gelişen sepsis, cerrahi açıdan önde gelen ölüm sebeplerinden biri olması nedeniyle ciddiyetini günümüzde de sürdürmektedir. Bu deneysel çalışma peritnit ve sepsisin patogenezinde önemli rol oynadığı kabul edilen serbest radikaller ile radikal tutucu ajanlardan alfa-tokoferol (AT) ve N-asetil sistein'in (NAC) sağkalım üzerindeki etkilerinin incelenmesi amaçlandı. Ortalama 210 gr ağırlığında 25 adet sağlıklı Wistar Albino cinsi erkek sıçan 5'er adetlik: I) kontrol (peritonit), II) peritonit öncesi alfa-tokoferol, III) peritonit öncesi a-tokoferol+N-asetil sistein, IV) peritonit sonrası alfa-tokoferol, V) peritonit sonrası alfa-tokoferol+N-asetil sistein gruplarına ayrılmışlardır. Peritonit canlı E.coli, her deneğe 6.6x109 suş şeklinde intraperitoneal olarak injekte edilmek suretiyle oluşturuldu. Her grup için peritonit sonrası 24 saatlik sağkalım oranları, periton dokusunda doku hasarını gösteren malondialdehid ve ksantin oksidaz düzeyleri, antioksidan enzim aktivitesini gösteren katalaz, süperoksid dismutaz (SOD) periton dokusunda; kanda ve periton dokusunda glutatyon peroksidaz (Gpx) düzeyleri ölçüldü. Abstract
Bacterial peritonitis and sepsis due to peritonitis are serious disease processes that produce substantial morbidity and mortality despite therapeutic interventions. Abundant evidence has implicated a number of reactive free radicals as sources of tissue injury inflammation. In this present study, the effect of alpha-tocopherol (AT) and N-acetyl cistein (NAC), which are free radical scavengers on the survival in peritonitis and sepsis was investigated. 25 Wistar Albino male rats with an average weight of 210 gr were divided into 5 groups; I) Control (peritonitis), II) alfa-tocoferol given before peritonitis, III) alfa-tocopherol and N-acetyl cistein given before peritonitis, IV) alfa-tocopherol given after peritonitis, V) alfa-tocoferol and N-acetyl cistein given after peritonitis. Peritonitis was induced by intraperitoneal injection of 6.6x109 live E. coli bacteria. For each group 24 hour survival rates, malondialdehyde and xantinoxidase levels which show damage in peritoneal tissue, catalase and superoxide dismutase (SOD) levels in periton tissue which show antioxidant enzyme activity, and glutation peroxidase (Gpx) levels in both blood and peritoneal tissue were measured and noted.
