PİNOT NOİR ÜZÜM ÇEŞİDİNDE SPONTAN MUTASYON OLUŞUMUNUN MORFOLOJİK VE MOLEKÜLER OLARAK İNCELENMESİ

Özet
Islah çalışmalarında mutajen kullanılarak uyarılmış mutasyon uygulamaları yürütüldüğü gibi, doğada daender olarak spontan mutasyonlar görülebilmektedir. İster uyarılmış ister spontan mutasyon sürecindetüm hücre tabakaları, gen bölgeleri aynı değişime uğramayarak aynı doku üzerinde morfolojik olarakmutant ve mutant olmayan farklı dokuların görüldüğü kimeralar oluşabilmektedir. Her iki durumu içinealabilecek benzer bir durum, arazi çalışmaları esnasında Tekirdağ’ın Şarköy İlçesindeki Pinot noir üzümçeşidi ile tesis edilmiş üretici bağında tespit edilmiştir. Aynı omca üzerinde farklı tane rengine sahipsalkımların olduğu bu durum morfolojik ve moleküler olarak incelenmiş, tane kabuk renginin değiştiğimutant genotip ile orijinal Pinot noir çeşidi kıyaslanmıştır. Morfolojik farklılıkları ortaya koymak adınaOIV tanımlayıcı skalası kullanılarak 24 ampelografik karakter üzerinde değerlendirmeler yapılmış,değerlendirme sonucunda başta tane kabuk rengi olmak üzere bazı karakterlerde farklılıklar tespitedilmiştir. OIV tanımlayıcı skalasında bulunan 6 SSR (Simple Sequnce Loci) lokusu ile yapılanmoleküler tanı analizinde ise iki örnek arasında mutasyon polimorfizm tespit edilememiştir. Ayrıca ikiörnek, asmada renk karakterini kontrol eden Myb ilişkili gen grubu açısından da değerlendirilmiştir.Analizler ve mevcut literatürler ışığında, oluşan mutasyonun Myb ilişkili gen grubu kaynaklı olmadığısaptanmıştır.
Abstract
Mutagenic mutations have been carried out in breeding studies, as well as spontaneous mutations rarely seen in nature. Both in the stimulated mutation and in the spontaneous mutation process, all cell layers and gene regions can not undergo the same variation. The presence of mutant and non–mutant parts on the same tissue is called chimera. A similar situation that could include both cases was determined in a vineyard established with Pinot noir grape variety in Şarköy district of Tekirdağ during the field studies. This situation that the bunches with different colored berries on the same vine are examined morphologically and molecularly. The original Pinot noir variety was compared with the mutant Pinot noir. In order to demonstrate the morphological differences, 24 ampelographic characters were evaluated using the OIV descriptive scale. As a result of the morphological evaluation, differences were found in some other characters as well as berry color. In the molecular diagnostic analysis performed with 6 SSR (Simple Sequnce Loci) loci on the OIV descriptor scale, mutation polymorphism could not be detected between two samples. In addition, two samples were evaluated for the gene group associated with Myb, which controls color characteristics in grape. According to the analyzes and available literature, it was determined that the mutation formed was not caused by the Myb related gene group.
