Beyaz Peynirde Nisini Tolere Eden Laktik Asit Bakteri Miktarı ve Çeşitliliği

Özet

Bu çalışmanın amacı, beyaz peynir florasından nisini tolere eden Laktik Asit Bakteri'nin (LAB) izolasyonu, takiben 16S rDNA dizi analizine göre tanımlanmasıdır. Denizli semt pazarlarından temin edilen beyaz peynirlerin mikrobiyolojik kalitesinin, çeşitli ulusal markalar altında ambalajlı olarak satılan peynirlerden oldukça düşük bulunmuştur. Bunun dışında beyaz peynirlerde M17 ve MRS ortamında gelişen LAB sayıları sırasıyla ortalama 8.38 ve 7.28 log kob/g olarak tespit edilmiştir. Aynı peynirlerde 500 IU/mL nisin katkılı M17 ve MRS ortamlarında gelişen LAB sayısı ise sırasıyla ortalama 4.99 ve 2.92 log kob/g olarak belirlenmiştir. Seçilen 23 adet nisini tolere eden LAB izolat; Lactobacillus plantarum (4), Lactobacillus casei (3), Lactobacillus paracasei (3), Lactobacillus pentosus (3), Pediococcus acidilactici (3), Enterococcus faecium (3), Lactobacillus rhamnosus (2), ve Lactobacillus fermentum (2) ile %97'nin üzerinde benzer bulunmuştur. Sonuç olarak, peynir florasında bulunan laktik suşlarının nisinden önemli seviyede etkilendikleri belirlenmiştir. Peynir üretimde nisin ile birlikte destek kültür olarak kullanılabilecek potansiyel laktik suşlar tanımlanmıştır.

Abstract

The aim of this study is the isolation of LAB tolerating nisin and identification within 16S rDNA sequencing ultimately. The microbiological quality of cheeses obtained from various Denizli local bazaars were found lower than the samples packaged and sold with commercial brands. The LAB counts grown in M17 and MRS were 8.38 and 7.29 log cfu/g, respectively. When 500 IU/mL nisin was inoculated to the M17 and MRS, the LAB counts in cheese samples were averagely 4.99 and 2.92 log cfu/g respectively. The 23 collected LAB tolerating nisin was found 97% similar within; Lactobacillus plantarum (4), Lactobacillus casei (3), Lactobacillus paracasei (3), Enterococcus faceium (3), Lactobacillus rhamnosus (2) and Lactobacillus fermentum (2). In conclusion, lactic cultures existed in white-brined cheese flora was affected from nisin significantly and potential lactic cultures which would be useful for the production of White-brined cheese production together with nisin were collected and identified.
