Östrustaki inek serumunda bulunan östradiol-17 $\beta$ düzeylerinin inek oositlerinin in vitro maturasyon ve fertilizasyonuna etkisi

Özet
Bu çalışmada, mezbahadan toplanan inek oositlerinin maturasyon ve fertilizasyonuna; östradiol 17-$\beta$ düzeyi belirlenmiş olan östrustaki inek serumu (ECS) etkisinin, FCS ile karşılaştırılması amaçlanmıştır. Siyah Alaca düvelerden östruslarının ilk 3 saati içerisinde alınan kanların serumları ile bu serumlardan hazırlanan stok serum bileşiminin ve FCS'nin içerdiği östradiol-17$\beta$ düzeyleri, RIA yöntemiyle belirlenmiştir. Morfolojik kalite sınıflamasına göre A ve B kalite olan inek oositleri kullanılmıştır. Maturasyon ortamlarında, %5, %10 ve %20 oranlarında olacak şekilde stok serum; kontrol grubunda ise aynı oranlarda FCS kullanılmış ve veriler varyans analizi kullanılarak test edilmiştir. Çalışma sonucunda düvelerin serumlarında sırasıyla; Düvel=18.4 pg/ml, Düve2=20.1 pg/ml, Düve3=21.8 pg/ml; stok serum bileşiminde ise 19.9 pg/ml; FCS içerisinde de, 34.8 pg/ml östradiol-17$\beta$ (E2) düzeyi tespit edilmiştir. Grup 1'de, %5, %10 ve %20 serum katılım düzeylerine göre sırasıyla, 0.78±0.$010^a$, 0.84±0.$009^b$, 0.86±0.$012^b$ maturasyon oranına ulaşılmış (p<0.01); Grup 2'de ise sırasıyla, 0.77±0.$010^a$, 0.82±0.$008^b$, 0.85±0.$010^b$ maturasyon (p<0.01); 0.51±0.$020^a$, 0.51±0.$023^a$, 0.59±0.$017^b$ fertilizasyon (p<0.01) oranına ulaşılmıştır. Sonuç olarak, östrustaki düvelerden elde edilen serumların (ECS) inek oositlerinin in vitro maturasyonunda ekonomik bir şekilde kullanılabileceği, %10 ve %20 düzeylerinde katılan ECS ve FCS'nin %5 düzeyine göre daha yüksek maturasyon ve fertilizasyon sonucu verdiği görülmüştür.
Abstract
The aim of this study was to compare influence of Estradiol-17$\beta$ levels measured in estrous cow sera (ECS) on oocytes from slaughtered in vitro maturation and fertilization with fetal calf sera (FCS). Three Holstein Fresian heifers were used for source of the sera. Blood was collected in the first 3 hours of the estrous from heifers and sera was decomposed from the blood then sera were mixed and stock serum was obtained. Level of estradiol-17$\beta$ in all the sera was analized by Radio Immuno Assay. Oocytes were selected and used as A and B quality grade morphologically. Stock serum was added in the maturation media 5%, 10% and 20%. The same proportions of FCS were added in the control group. Data were tested by ANOVA. Estradiol-17$\beta$ (E2) levels in the heifers blood sample were found for Heiferl, Heifer2 and HeiferS and stock serum as respectively; 18.4 pg/ml, 20.1 pg/ml, 21.8 pg/ml and 19.9 pg/ml. Estradiol 17-$\beta$ was also measured 34.8 pg/ml in the FCS. For adding 5%, 10% and 20% proportions of ECS stock serum, the results were respectively; 0.78±0.$010^a$, 0.84±0.$009^b$, 0.86±0.$012^b$ maturation (p<0.01). The results of addition of FCS as a control group for 5%, 10% and 20% sera levels were 0.77±0.$010^a$, 0.82±0.$008^b$ 0.85±0.$010^b$ maturation (p<0.01); 0.51±0.$020^a$, 0.51±0.023^a,$ 0.59±0.017b fertilization result As a result, estrous cow serum can be used economically for in vitro cow oocytes maturation. When ECS and FCS 10% and 20% were added in the maturation media, more better maturation and fertilization results were found according to 5% ECS and FCS.
