Diyabetik Ratlarda Zakkum (Nerium oleander L.) Çiçeği Etanolik Liyofilize Ekstresinin Eritrosit Frajilite, Hematolojik ve Antioksidan Etkilerinin Araştırılmas

Özet

Bu çalışma streptozotosin (50 mg kg-1) ile Tip 1 diyabet oluşturulanratlarda zakkum (N. oleander) çiçeğinden elde edilen etanolikliyofilize ekstresinin eritrosit frajilite, hematolojik parametreler, totalantioksidan kapasite (TAS), total oksidan kapasite (TOS) ve oksidatifstress indeksi (OSI) üzerine olan etkilerine odaklandı. Toksisite testisonrası, 49 rat 7 gruba ayrıldı. Kontrol, Nerium oleander (NO) (50 mgkg1), Diabetes mellitus (DM), DM+Gly-3 (3 mg kg1), DM+NO-25 (25mg kg1), DM+NO-75 (75 mg kg1) ve DM+NO-225 (225 mg kg1). 21günlük uygulamadan sonra elde edilen bulgulara göre, DM grubundaeritrosit hemolizi (%0.2-0.6 NaCI konsantrasyonlarda) kontrolgruplarına göre önemli artış gösterirken (P<0.05); özellikle DM+NO225 grubunda eritrosit hemolizi önemli oranda azaldı (P<0.05).Hematolojik parametreler, bazı gruplarda dalgalanmalargöstermesine rağmen, DM+Gly grubunda eritrosit (RBC), hemoglobin(HGB) ve hematokrit (HCT) düzeyleri diğer gruplara göre önemli artışgösterdi (P<0.05). Ayrıca, DM grubu lökosit (WBC), eritrosit dağılımgenişliği (RDW) ve ortalama eritrosit hacmi (MCV) değerleri kontrolgruplarına göre önemli artış gösterirken, eritrosit ortalamahemoglobin konsantrasyonu (MCHC) ise önemli oranda azaldı(P<0.05). DM grubunda plazma TAS seviyesindeki azalma, ile TOS veOSI değerindeki artış kontrol ve uygulama gruplarına göre önemlibulundu. N. oleander çiçek liyofilize ekstresinin STZ ile diyabetoluşturulan ratlarda diyabetin sebep olduğu artan hemoliziönleyici/azaltıcı etkilere ve antioksidan özelliklere sahip olabileceğisonucuna varılmıştır.

Abstract

The present study was focused on the erythrocyte fragility, hematological parameters, total antioxidant capacity (TAS), total oxidant capacity (TOS) and oxidative stress index (OSI) of flower ethanolic lyophilized extract obtained from N. oleander plant on Streptozotocin (50 mg kg-1)-induced Type 1 diabetes in rats. After toxicity test, 49 rats were divided into 7 groups. Control, Nerium oleander (NO) (50 mg kg-1), Diabetes mellitus (DM), DM+Gly-3 (3 mg kg-1), DM+NO-25 (25 mg kg-1), DM+NO-75 (75 mg kg-1) and DM+NO225 (225 mg kg-1). Based on the results, after 21 days of application, erythrocyte hemolysis of DM group was increased (at 0.2-0.6% NaCI concentrations) compared to Control groups whereas treatment groups significantly decreased (P<0.05) erythrocyte hemolysis, especially in DM+NO-225 group. Although, hematological parameters showed some fluctuations in the groups, DM+Gly group showed significant increase (P<0.05) in erythrocyte (RBCs), hemoglobin (HGB), hematocrit (HCT) levels compared to all other groups. In addition, leukocyte (WBC), red cell distribution width (RDWs) and mean corpuscular volume (MCV) values of DM group increased with respect to the control groups whereas mean cell corpuscular
