Ratlarda Deneysel Akciğer Fibrozisinde Nitrik Oksit Oksidasyon Ürünleri ve Kan Gazları Düzeylerinin Araştırılması *

Özet
Bu araştırmada, antitümoral etkili bir ajan olan bleomisinin yan etkisi olarak şekillenen akciğer fibrozisinin, nitrik oksit metabolitleri ve kan gazları seviyeleri üzerine olan etkileri araştırıldı. Hayvan materyali olarak 16 Wistar-Albino ırkı erkek rat kullanıldı. Akciğer fibrozisini oluşturmak için bleomisin hidroklorid 7.5 mg/kg oranında tek doz intratrakeal olarak kullanıldı. Yapılan analizlerde, istatiksel açıdan kontrol grubuna göre nitrit seviyesindeki artışlar p 0.05, tCO2 düzeyindeki artışlar p 0.05). Histopatolojik bulgularda akciğer fibrozisinin oluşumu tespit edilmiştir. Sonuç olarak bleomisinin sebep olduğu akciğer fibrozisinde hücrede meydana gelen hasardan ve oksidatif stresten dolayı kan gazları seviyeleri düşmüş ve nitrik oksit metabolitlerinde artış gözlenmiştir.
Abstract
The levels of nitric oxide oxidation products and blood gases were studied on the experimental pulmonary fibrosis induced by a side effect of bleomycine, an antitumoral agent in rats. In this study 16 Wistar-Albino rats were used. The same animals values prior to the experimental treatment were considered as the control. To establish the experimental fibrosis, bleomycine hydrochloride was used intratraceally at a single dose of 7.5 mg/kg. After the fibrosis was performed, nitrite, nitrate and blood gases were carried out in the blood and serum. From the analysis made, the statistical are; interms of the control group; increase in the nitrite rate is p<0.001, the increases in the nitrate rate is p<0.001, the decreases in pCO2 is p<0.001, the increases in pH is at rate of p<0.001, the decreases in pO2 isp>0.05 and the increase at the rate of tCO2 is p<0.05. These findings have great importance. At the rate of HCO3 no statistical difference was found (p>0.05). Formation of the pulmonary fibrosis was demonstrated by histopathology findings. As a result, in lung fibrosis, which is caused by bleomycin, the changes in rates of nitric oxide oxidation products and blood gases show that there may have been oxidative stress, and it means that fibrosis has occurred in lung.
