Gebeliğin son döneminde verilen etanolün yavru ratların duktus epididimis ve testis dokusu üzerine etkisi

Özet
Bu deneysel galismada, hamileligin son donemlerinde verilen etanolun dogan yavru ratlarda gelisme geriligine rieden olup olmadigimn, testislerde spermatogenezisi ve spermatik hucreleri etkileyip etkilemediginin histopatolojik olarak incelenmesi amaglanmistir. Qalismada, 12 adet Wistar albino cinsi gebe rat kullanildi. Deney grubu (n= 6) ratlara, gebeligin 17'nci gununden 21'inci gune kadar hergiin 0.5 ml % 96'hk etanol + 0.5 ml serum fizyolojik intraperitoneal olarak enjekte edildi. Dogumdd|n sonra, deney grubu (n= 17) ve kontrol grubu (n= 14) erkek yavrulann vucut agirhklan olgulerek testisleri eter ahestezisi altinda gikanldi ve % 10'luk formaline ahhdi. Rutin histolojik i§lemlerden sonra kesitler, hematoksilen-eozin ile boyanip isik mikroskobunda incelendi. Etanol grubu yenidogan ratlann toplam vucut agirhklan ortalamasi, kontrol grubu ratlannkinden anlamli olarak daha dusuk bulundu (p< 0.001). Johnsen testikuler biyopsi skorlamasina gore, etanol grubunda ortalama histolojik hasarlanma grade 5.6+0.8 duzeyinde iken, kontrol grubunda ortalama 8.8+1.2 olarak bulundu (p< 0.001). Etanol grubunda, kontrol grubuna gore testislerde goreceli olarak 11.8 kat daha fazla histopatolojik hasarlanma oldugu belirlendi (p= 0.004). Bu galismada, etanolun etkisiyle testislerdeki tubulus yapisinm normal yapiya gore degistigi, hucrelerde hafif derecede dejeneratif degisikliklerin oldugu, tiibuller arasi bag dokusu ve Leydig hucrelerinin etkilendigi gozlenmistir. Sonug olarak, etanolun gebelik doneminde kullanildigmda dogan yavru ratlann toplam vucut agirligini dusurerek gelisme geriligine neden oldugu ve ayrica testislerin histolojik yapisinda dejenerasyon yaptigi, ancak duktus epididimisin yapisinda histopatolojik olarak anlamli bir dejenerasyon yapmadigi anlasilmistir.
Abstract
Histopathological Changes Occuring In Ductuli Epididymis And Testes Of Litters When Pregnant Rats Are Exposed To Ethanol Late During Pregnancy-In this experimental study, we aimed to evaluate whether ethanol effects total body growth, spermatogenesis and sperm cells when mothers were exposed during late periods of pregnancy. Twelve pregnant Wistar Albino rats were used in the study. A mixture of 0.5 ml 96 % ethanol + 0.5 ml % 0.9 NaCl was intraperitoneally injected every day between days 17 and 21 of pregnancy in the study group (n= 6). After birth, orchiectomies were, done under'ether anesthesia in both controls (n= 14) and experimental males (n= 17). The tissues were fixed in 10 % formaline. After routine histological process, the sections were stained with hematoxylene-eosin and evaluated under light microscopy. The mean body weight of newborn rats exposed to ethanol was significantly lower, when compared to that of controls (p< 0.001). According to Johnsen's testicidar biopsy scoring, the mean grade of histopathological damage in the ethanol group ivas noted as 5.6± 0.8, whereas the mean score was 8.8 ± 1.2 in the control group (p< 0.001). The tissue damage in the ethanol group was 11.8 times greater than that in the control group (p- 0.004). It is evident in this study that ethanol caused changes in the structure of testicidar tubidi, mild degenerative changes in cellular structure, intertiibular connective tissue and Leydig cells when pregnant rats were exposed during pregnancy. In conclusion, if rats are exposed during pregnancy, ethanol causes decrease in body weight that appear as growth retardation in the newborns, and also significant degeneration in histological structure of the testes. However, the epididymal ducts are spared of these effects.
