Fasulye fidelerinin (PHaseolus vulgaris L. ev. Strike) kök, gövde ve yaprak büyümesi üzerine nikel $(Ni^{+2})$ ve Krom' un $(Cr^{+3})$' un etkileri

Özet
Bu çalışmada fasulye fidelerinin kök, gövde ve yaprak büyümesi üzerine nikel $(NiCl_2.6H_2O)$ ve krom $(CrCl_3.6H_2O)$ ’un etkileri araştırılmıştır. Çalışmada bir haftalık fasulye fideleri kullanılmıştır. Her iki metalinde klor tuzu kullanılmıştır. Ağır metal solüsyonları saf suyla hazırlanmıştır. Nikelin stok solüsyonundan $(NiCl_2.6H_2O)$ 0.5, 0.7, 1.0 mM ve kromun stok solüsyonundan $(CrCl_3.6H_2O)$ ise 0.5, 0.7, 1.0 mM konsantrasyonları hazırlanmıştır. Fideler on gün süreyle hoagland solüsyonuyla hazırlanmış nikel (0.1, 0.3, 0.5 mM) ve krom (0.5, 0.7, 1.0 mM) klorürün farklı konsantrasyonlarında büyümeye bırakılmıştır. Fidelerin her gün kök ve gövde uzunlukları ile yaprak alanları ölçülmüştür. Elde edilen sonuçlara göre her iki ağır metalin de fidelerin kök, gövde ve yaprak büyümesini önemli oranlarda engellediği tespit edilmiştir. Bu ağır metallerin konsantrasyonlarındaki artış ile kök, gövde ve yaprak büyümesinin inhibisyon oranı arasında bir paralellik görülmüştür. Fidelerin ağır metale maruz kalma süresinin uzaması kök, gövde ve yaprak büyümesindeki azalmanın daha fazla olmasına yol açmıştır. Ayrıca kök, gövde ve yaprak büyümesindeki azalışın ağır metalin çeşidi ve konsantrasyonuyla ilişkili olduğu görülmüştür. Nikel ve krom stresine kök büyümesinin daha duyarlı olduğu, bunu gövde ve yaprak büyümesinin takip ettiği belirlenmiştir. Bu iki ağır metalden nikelin kroma göre daha toksik olduğu tespit edilmiştir. Sonuçlar istatistik açıdan P<0.05 ve P<0.01 düzeylerinde anlamlı bulunmuştur.
Abstract
In this study, the negative effects of nickel $(NiCl_2.6H_2O)$ and chromium $(CrCl_3.6H_2O)$ treatments on the root shoot and leaf growth of the bean seedling were investigated. Also one week old bean seedlings were used. The chlorine salts of each two heavy metals were used. Heavy metal solution prepared with pure water. The stock solution of nickel was prepared at a concentration of 0.5, 0.7, 1.0 mM $(NiCl_2.6H_2O)$ and other stock solution had a concentration of 0.5, 0.7, 1.0 mM for chromium (0.5, 0.7, 1.0 mM). Seedlings were left in the nickel and chromium chloride in different concentration prepared with hoagland solution growing. The elongation of seedling root, shoot and area of leaves were measured daily. According to the obtained results, both heavy metals significantly prevented the growth of root, shoot and leaves of seedlings. A parallel correlation was observed between increases in the concentration of these heavy metals and inhibition of root, shoot and leaf growth. Extending of exposure time to heavy metals of seedling lead to decreasing of root, shoot and leaf growth. It was found that the decreases in the root shoot and leaf growth related with kind of the heavy metal and concentrations. It was determined that nickel and chromium stress is more sensitive the root growths, followed shoot and leaf growth, respectively. It was determined that each other nickel and chromium to compare, nickel had the highest toxic effect. The results were found to be meaningful significant (P<0.05 and P<0.01).
