Kafeik Asit Fenetil Ester Sıçanlarda Göğüs Travması Sonrası Gelişen Hepatik Hasarı Nükleer Faktör Kappa Beta ve İndüklenebilir Nitrik Oksit Sentezini Baskılayarak Azaltır

Özet
Amaç: Bu çalışmada göğüs travması modeli oluşturulan sıçanlarda meydana gelen hepatik hasara karşı kafeik asit fenetil ester (KAFE)' nin Nükleer Faktör Kappa Beta (Nf-κβ) ve indüklenebilir nitrik oksit sentaz (iNOS) aktivitesi üzerine etkisinin incelenmesi amaçlandı. Yöntem: Çalışmada 40 adet erişkin Wistar albino erkek sıçan sırasıyla 4 gruba ayrıldı; kontrol, göğüs travma modeli, göğüs travma modeli+KAFE ve KAFE grubu. KAFE uygulaması 7 gün boyunca 10 µmol/kg/gün dozda intraperitoneal olarak gerçekleştirildi. Deney süresi sonunda karaciğer dokusu alınarak meydana gelen histopatolojik değişiklikler Hematoksilen-Eozin (H&E) boyaması ile değerlendirildi. Ayrıca hepatik Nf-κβ ve iNOS aktiviteleri indirek immünohistokimyasal yöntemle incelendi. Bulgular: Göğüs travması uygulanan grupta hepatosit kordonlarında dejenerasyonları ve sinüzoidal dilatasyon bulgularına rastlandı. Göğüs travması+KAFE grubunda histopatolojik bulguların göğüs travması grubuna göre daha hafif seyrettiği belirlendi. Ayrıca, göğüs travması+KAFE grubunda, göğüs travması grubuna kıyasla hepatik Nf-κβ ve iNOS immünreaktivitesinde istatistiksel olarak anlamlı derecede bir azalma olduğu görüldü (p<0.05). Sonuç: KAFE' nin göğüs travması uyarımlı sıçan hepatik hasarında Nf-κβ ve iNOS aktivitesini baskılayarak koruyucu etki gösterdiği belirlendi.
Abstract
Objective: The aim of this study was to investigate the effect of caffeic acid phenethyl ester (CAPE) on Nuclear Factor Kappa Beta (Nf-κβ) and inducible nitric oxide synthase (iNOS) activity against hepatic injury with chest trauma model in rats. Method: In the study, 40 adult Wistar albino male rats were divided into 4 groups respectively; control, chest trauma, chest trauma+CAPE and CAPE groups. CAPE administration was performed intraperitoneally at a dose of 10 μmol / kg / day for 7 days. At the end of the experiment, histopathologic changes were determined by hematoxylin-eosin (H&E) stain in liver tissue. In addition, hepatic Nf-κβ and iNOS activities were examined by indirect immunohistochemical method. Results: Degeneration of hepatocyte cords and sinusoidal dilatation findings were found in the chest trauma group. Histopathological findings of chest trauma + CAPE group were found to be milder than chest trauma group. In addition, a statistically significant reduction in hepatic Nf-κβ and iNOS immunoreactivity was seen in the chest trauma + CAPE group compared to the chest trauma group (p<0.05). Conclusion: CAPE has been shown to exert protective effects by suppressing Nf-κβ and iNOS activity in chest trauma induced hepatic damage in rats.
