Kolesterol ve sıvı yağ ilave edilmiş diyetle beslenen ratlara vitamin C ilavesinin antioksidan ve plazma lipid profiline etkilerinin değerlendirilmesi

Özet
Bu çalışmada; rat rasyonlarına zeytinyağı (%5), kolesterol (%0.5) ve vitamin C (500 mg/kg yem) ilavelerinin plazma MDA, total kolesterol, HDL-LDL kolesterol, trigiliserit, eritrosit GSH, karaciğer ve beyin dokusu MDA ve GSH konsantrasyonlarına etkileri araştırıldı. Araştırmada 6 aylık, canlı ağırlıkları birbirine yakın, toplam 50 adet Sprague-Dawley erkek rat kullanıldı. Bu hayvanlar herbirinde 10 adet fat içeren 5 gruba ayrıldı. Araştırma Selçuk Üniversitesi Veteriner Fakültesi Deney Hayvanları Ünitesi'nde yürütüldü ve 60 gün devam etti. Çalışmada kontrol (l.grup), kolesterol (2.grup), zeytinyağı (3.grup), kolesterol + vitamin C (4.grup), zeytinyağı + vitamin C (5.grup) grupları oluşturuldu. Deneme sonunda kardiyak punksiyonla alınan kan örneklerinden plazma, serum ve eritrosit süspansiyonları elde edilerek plazma MDA, kolesterol, HDL-LDL kolesterol, trigliserit, eritrosit GSH, ile karaciğer ve beyin MDA, GSH ölçümleri yapıldı. Ratlarda deneysel olarak yürütülen bu çalışmada zeytinyağı ve kolesterol ilaveleri kan lipid profilinde belirgin farklılıklar ortaya çıkarmış, zeytinyağı verilen grupta iyi huylu kolesterol (HDL) düzeyleri yükselmiş ve LDL kolesterol düzeyleri kolesterol grubuna oranla belirgin düşüş göstermiştir. Vitamin C katkısı ile bu düşüş daha belirgin olarak ortaya çıkmıştır? Plazma MDA değerleri de lipid peroksidasyonunun en yüksek zeytinyağı grubunda şekillendiğini göstermektedir. Beyin MDA değerlerinin plazma MDA ile uyumlu şekilde zeytinyağı grubunda en yüksek olduğu saptanmıştır. Sonuç olarak, ratlarda yürütülen bu çalışmada, vitamin C uygulamasının özellikle doymamış yağ asitlerinin lipid peroksidasyonuna duyarlılıklarını azaltabileceği ve kan lipid profiline düzenleyici etkileri olabildiği kanısına varılmıştır.
Abstract
In this study, the effects of supplementation of cholesterol and vitamin C (250 mg/kg feed) on the erytrocyte GSH, plasma MDA, total cholesterol, HDL-LDL cholesterol and triglyceride concentrations, brain and liver malondialdehyde (MDA), glutathione (GSH) levels of rats were investigated! A total of 50 Sprague- Dawley male rats which have 6 months age, in equal of body weight were used. During the experiment, these animals fed with standart ration as ad libitum. Animals were housed in S.Ü.Veterinary Faculty Experimental Animals Unit. Experimental period was lasted at 60. day. There were five experimental groups in this study as following; 1. Control, 2. Cholesterol (% 0.5), 3. Olive oil.(% 5), 4. Cholesterol plus vitamin C (500 mg/kg), 5. Olive oil plus vitamin C (500 mg/kg). At the end of the experiment, blood samples were taken by cardiac puncture and erytrocyte GSH, plasma MDA, cholesterol, HDL-LDL cholesterol, triglyceride and also GSH and MDA concentrations in brain and liver tissue of rats were determined. Supplementation of olive oil and cholesterol into rat diets made significant differences in lipid parameters, also HDL cholesterol levels were increased in olive oil group and LDL cholesterol was lower than the cholesterol fed group. Moreover, these decrease in LDL and triglyceride levels were more significant with plus vitamin C supplementation. The high plasma MDA levels have shown that lipid peroxidation was highly occurred in olive oil added group and also the highest brain MDA levels was determined in olive oil fed group. As a result^ the findings of the presenting study suggest that vitamin C addition may decrease the sensjjtivities of unsaturated fatty acids against to oxidation. It was also observed that blood lipid parameters were regulated by vitamin C supplementation in rats.
