Özet
İneklerde korpus luteum'dan (CL) progesteron salgılanmasının devamlılığı gebeliğin oluşmasında zorunludur. İneklerde luteolitik prostaglandin $F_{2alpha} (PGF_{2alpha})$ salınımı, gebeliğin erken döneminde CL regresyonuna ve progesteron salınımının kesilmesine sebep olur. Uterus içerisinde sağlıklı bir embriyo olması halinde gebeliğin oluşması ve devamlılığı için ilk olarak embriyo ve uterus ile CL arasında bir takım kompleks mekanizmaların devreye girmesi gerekmektedir. Bu kompleks mekanizma embriyonun trofoblast hücrelerinden salgılanan proteinler vasıtası ile başlatılır. Bu spesifik trofoblast proteinlerinden birisinin interferon proteinleri ile homolog olduğu tespit edilmiş ve daha sonra Interferon-tau $(IFN-tau)$ olarak tanımlanmıştır, ineklerde $IFN-tau$ 'unun fonksiyonlarından bir tanesi ve en önemlisi endometriumdan $(PGF_{2alpha})$'nın pulsatif salmımmı durdurarak CL regresyonunu engellemektir. IFN-t'unun $(PGF_{2alpha})$ salmımmı durdurduğu in vitro olarak inek endometrium hücre kültürlerinde gösterilmiştir. Son yıllarda, $IFN-tau$ 'unun moleküler düzeyde luteolitik $(PGF_{2alpha})$ 'nın salınımını nasıl durduğu yoğun bir şekilde araştırılmaktadır. Yapılan çalışmalar; $IFN-tau$ tarafından başlatılan çeşitli düzenleyici mekanizmaların varlığını ortaya koymuştur. Günümüzdeki bilgilere göre, $IFN-tau$'nun endometriumda epitel hücrelerinde doğrudan $(PGF_{2alpha})$ salınımını düzenleyen östrojen reseptörü alfa (ERa) ve ok-sitosin reseptörü (OTR) ekspresyonunu azalttığı yönündedir. IFN-x'nun endometrial epitel hücrelerinde prostaglandin H sentaz-2 (PGHS-2) geni üzerine etkisi de alternatif bir mekanizma olarak tartışılmaktadır.
Abstract
Maintenance of secretion of progesterone from the ovarian corpus luteum (CL) is essential for establishment of pregnancy in cows. The luteolytic prostaglandin (PG) is $(PGF_{2alpha})$ in cows causing regression of the corpus luteum and cessation of progesterone production. Pregnancy is established and maintained through a complex interaction between the conceptus and the maternal environment including the uterus and corpus luteum. This complex interaction was found to be driven by trophoblastic proteins. A specific trophoblastic protein was determined to have homology with interferons and later identified as interferon-tau ( $IFN-tau$ ). One function of $IFN-tau$ is to extend the lifespan of corpus luteum and to reduce the pulsatile release of $(PGF_{2alpha})$ from the endometrium in cows. A clear inhibition of secretion of $(PGF_{2alpha})$ by IFN-x in vitro from bovine endometrial cells has been shown by a number of studies. However, the cellular mechanisms as to how $(PGF_{2alpha})$ secretion is inhibited by trophoblastic $IFN-tau$ have been under extensive investigation. Studies over the years have suggested various regulatory mechanisms as to how $IFN-tau$ exerts its effect to reduce $(PGF_{2alpha})$ secretion to maintain the corpus luteum. Current explanations are that $IFN-tau$ acts on the endometrium to reduce expression of estrogen receptor alpha (ERa) and oxytocin receptor (OTR) that are involved directly in stimulating pulsatile $(PGF_{2alpha})$ secretion from uterine epithelial cells. Alternatively, $IFN-tau$ acts on uterine epithelial cells to cause transcriptional inactivation of the PG H Synthase-2 (PGHS-2) gene.
