Deneysel Diyabet Oluşturulan Ratlarda Prooksidan / Total Antioksidan Durumu ve Vitamin Düzeyleri Üzerine Likopenin Etkisi

Özet
Bu çalışma, deneysel olarak diyabet oluşturulan ratlarda antioksidan olarak koruyuculuğu bilinen likopen uygulanmasının TBARS, TAS ve vitamin (α-tokoferol, retinol, D3) seviyeleri üzerine etkilerini ortaya koymak amacıyla planlandı. Diyabet oluşturmak için D ve D+L grubundaki ratlara 45 mg/kg tek doz streptozotosin (STZ) intraperitoneal (i.p.) uygulandı. Kan şekerleri 270 mg/dL ve üzerinde olanlar diyabetik olarak kabul edilip çalışmaya dahil edildi. L ve D+L grubundaki ratlara likopen ayçiçeği yağında çözdürülerek 10 mg/kg/gün olarak uygulandı. Dört haftalık deneme süresinden sonra, kontrol ve likopen gruplarına göre diyabet grubunda TBARS, HbA1c ve glukoz seviyesinin arttığı, buna karşın TAS, retinol ve α-tokoferol düzeylerinin azaldığı tespit edildi (P<0.001). Diyabet grubuna göre diyabet oluşturulup likopen verilen grupta istatistiksel açıdan önem arz etmeyecek şekilde (P>0.05) TBARS seviyesinin düştüğü, TAS ve α-tokoferol seviyesinin yükseldiği, HbA1c ve glukoz seviyesinin ise istatistiksel olarak önem gösterecek şekilde düştüğü (P<0.001) yine de kontrol grubundan yüksek olduğu, retinol seviyesinin kontrol grubu ile aynı olduğu bulundu. Vitamin D3 seviyeleri açısından gruplar arasında fark olmadığı saptandı (P>0.05). Elde edilen sonuçlar diyabetik kişilerin diyetlerinde likopen ihtiva eden gıdaları tüketmelerinin, diyabetin neden olduğu hücre hasarının önlenmesinde görev alan antioksidan sistemleri güçlendirebileceği kanısına varıldı.
Abstract
The aim of this study, lycopene known as antioxidant was applied to rats with experimental diabetes to determine the effects of TBARS, TAS and vitamins (α-tocopherol, retinol, D3) levels. In order to induce diabetes in the rats D and D+L groups were given 45 mg/kg single dose streptozotocin (STZ) intraperitoneally (i.p.); lycopene dissolved in sunflower oil was administered (10 mg/kg/day) to the rats in the L and D+L groups. Blood glucose rise 270 mg/dl those with diabetes can be considered to be enrolled in the study. After the four-week experiment, there was increase in the TBARS, HbA1c and glucose levels while in decrease TAS, retinol an α-tocopherol levels in diabetic group were significanly different compared to the control and lycopene groups (P<0.001). Accordig to diabetic group, in the lycopene+diabetic group the decrease in TBARS level and an increase in TAS and α-tocopherol levels were seen, these alternations were not sufficient to constitute a statistical importance, retinol levels in the lycopene+diabetic group were similar to control group. According to diabetic group, HbA1c glucose levels were decreased in the lycopene+diabetic group (P<0.001), but these levels were higher than control group. The results suggest that consuming food include lycopene in diets of diabetic humans may serve to reinforce the antioxidant systems, thus having a protective effect against cell damage by diabetes.
