Tavşanlarda endotoksinle oluşturulan uveitte propolisin etkinliği

AMAÇ: Endotoksinle oluşturulan uveit (EU) insanlardaki akut ön uveite benzeyen bir hayvan oküler inflamasyonun modelidir ve yeni antiinflamatuar tedavileri denemek için yararlıdır. Çalışmanın amacı, bu modelde doğal bir bal arısı ürünü olan propolisin antiinflamatuar etkisini incelemek ve kortikosteroidlerle karşılaştırmaktır. YÖNTEM: Tavşanların sağ gözlerine 2,000 ng Salmonella typhi endotoksini intravitreal verilerek EU oluşturuldu. Hayvanlar rastgele 3 eşit gruba ayrıldı: Grup A'ya (kontrol) tedavi verilmedi. Grup B'ye metilprednizolon (5 mg/0.1 ml) (pozitif kontrol), Grup C'ye propolis (5 mg/0.16 ml) ön subtenon olarak uveit indüksiyonu esnasında ve 4. ve 8. saatlerde verildi. İnflamasyon, klinik bulgular, aközdeki inflamatuar hücre sayısının, protein içeriğinin ve nitrit/nitrat konsantrasyonunun ölçülmesiyle değerlendirildi. BULGULAR: Klinik derecelendirme, aköz hümör hücre sayısı, protein ve nitrit/nitrat konsantrasyonları metilprednizolon (1.8±0.7; 572±137 hücre/µl; 15.2±1.8 mg/ml; 22.5±6.3 µM/ml) ve propolis (2.3±0.5; 503±124 hücre/µl; 13.8±1.5 mg/ml; 19.4±5.8 µM/ml) gruplarında, kontrol grubuna (6.0±0.8; 2519±470 hücre/µl; 32.9±2.4 mg/ml; 49.3±15.2 µM/ml) göre istatiksel olarak anlamlı seviyede düşüktü (p 0.05). SONUÇ: Propolis tavşanlarda oluşturulan EU'de belirgin antiinflamatuar etki göstermiştir. Etki mekanizması konusunda ileri çalışmalara ihtiyaç vardır.
Abstract
BACGROUND: Endotoxin-induced uveitis (EIU) is an animal model of ocular inflammation mimicking the human disease of acute anterior uveitis and is useful for testing new anti-inflammatory therapy. The purpose of this study was to test the anti-inflammatory effect of propolis, a natural bee product, and compare it with corticosteroids on EIU. METHODS: EIU was produced in rabbits by intravitreal injection of 2,000 ng Salmonella typhi endotoxin into right eyes. Animals were randomized in three equal groups as follows: Group A (control) received no treatment, Group B received metylprednisolone (5 mg/0.1 ml) (positive control), and Group C received propolis (5 mg/0.16ml) by anterior subtenon injection at the time of uveitis induction and at 4 and 8 hour after the induction. Inflammation was evaluated by clinical manifestations, and by measuring protein and nitrite/nitrate levels and inflammatory cell content in the aqueous humor. RESULTS: Aqueous humor clinical grade, cell count, protein and nitrite/nitrate levels were the following: control group (6.0±0.8; 2519±470 cells/µl; 32.9±2.4 mg/ml; 49.3±15.2 µM/ml); metylprednisolone group (1.8±0.7; 572±137 cells/µl; 15.2±1.8 mg/ml; 22.5±6.3 (µM/ml); and propolis group (2.3±0.5; 503±124 cells/µl; 13.8±1.5 mg/ml; 19.4±5.8 µM/ml). Statistical significant differences were achived by the treatment groups when compared to control group (p<0.001). The effects of metylprednisolone and propolis groups were similar (p>0.05). CONCLUSION: Propolis showed significant antiinflammatory effect in EIU in rabbits. The mechanisms of its action warrant further investigation.
