Çeşitli glokom ilaçlarının insan subkonjonktiva fibroblast çoğalmasına etkisi

Özet
Amaç: Güncel sık kullanılan bazı glokom ilaçlarının subkonjonktiva fibroblast hücre çoğalmasına etkisinin değerlendirilmesi. Gereç ve Yöntem: Beyin ölümü gerçekleşmiş donörlerden elde edilen insan subkonjonktiva dokuları, RPMI besi vasatları kullanılarak uygun laboratuar şartlarında kültüre edildi. Timolol maleat %0.5, latanoprost %0.005, brimonidin tartrat %0.1 ve dorzolamid hidroklorür %2 gibi sık kullanılan antiglokomatöz ajanlar beş ayrı konsantrasyonda ayrı ayrı besi yerlerine damlatıldı. Hiçbir damla damlatılmamış olan bir besi yeri ise kontol grubunu oluşturdu. Hücre kültürünün 8. gününde Tripan mavisi canlılık testleri kullanılarak fibroblast hücre çoğalmaları değerlendirildi. Bulgular: Latanoprost, Dorzolamid, timolol and brimonidinin lethal dozları sırasıyla 1.97, 2.23, 3.47 and 3.51mgr/ml olarak tespit edildi. Tüm ilaçlar hücre inhibisyonunu sağlamada kontrole göre anlamlı derecede daha etkindi (tüm ilaçlar için, p 0.05). Sonuç: Sık kullanılanılmakta olan güncel antiglokomatöz ajanlar subkonjonktiva fibroblast çoğalmasını etkin bir şekilde inhibe etmektedir. Bu durum, başta latanoprost olmak üzere bu tür ilaçları kullanan glokom hastalarında sonradan ihtiyaç duyulabilecek filtran cerrahilerinin başarı şansını olumlu yönde etkileyebilir.
Abstract
Purpose: To assess the effect of currently used various antiglaucomatous drugs on human subconjunctival fıbroblast cellular proliferation. Material and Methods: Human subconjunctival fibroblast tissue, which was obtained from donors who were already diagnosed as the brain death, has been cultured under suitable laboratory conditions using RPMI medium. Each of frequently used antiglaucomatous drugs namely timolol maleate 0.5%, latanoprost 0.005%, brimonidine tartrate 0.1% and dorzolamide HCl 2% which were prepared in five different concentrations were dropped to the separate cellular medium. Another cellular medium, where no drug was dropped, was regarded as a control group. Trypan blue tests were used for assesment of the cellular vividity at the 8th day of the cellular culture. Results: The lethal dose of the drugs were 1.97, 2.23, 3.47 and 3.51 mgr/ml for latanoprost dorzolamide, timolol and brimonidine, respectively. All the drugs were found to significantly inhibit the subconjunctival fibroblast proliferation with respect to the control (p<0.001, for all drugs). The highest celllular inhibition was detected for latanoprost (p<0.05) whereas the lowest cellular inhibition was for brimonidine (p<0.05). Dorzolamide and timolol were found to have similar inhibition on the fibroblasts (p>0.05). Conclusion: Frequently used antiglaucomatous drugs have been found to inhibit the subconjuntival fibroblasts significantly. This may positively effect the success of a future filtrating surgery in glaucoma patients who use these drugs, especially latanoprost.
