Effect of isoflurane treatment on antioxidant enzyme levels in guinea pig liver

Özet: Amaç: İzofluranın hepatotoksik etkisinin enzimatik antioksidan sistemle ilgisinin olup olmadığını araştırmak.Materyal ve Metod: Onbsş kobay üç gruba ayrıldı: I-kontrol, II-izofluran, III-oksijen. Oksijen grubundaki hayvanlara % 100 oksijen ve izofluran grubundaki hayvanlara ise oksijen içinde (% 100, 2lt/dk.) izofluran (% 2 v/v) günde 30 dk olmak üzere arka arkaya 3 gün verildi. Hayvanların karaciğer dokularında süperoksit dismutaz (SOD), katalaz (CAT) ve glutatyon peroksidaz (GSH-Px) enzim aktiviteleri ve malodialdehid seviyeleri ölçüldü. Ayrıca serum ALT ve AST seviyeleri de tayin edildi.Bulgular: İzofluran ve oksijen gruplarında, kontrol grubuna göre SOD ve CAT aktivitelerinin değişmediği, GSH-Px aktivitesinin ise azaldığı gözlendi. Kontrol grubuna göre, izofluran grubunda MDA seviyesinin değişmediği fakat, oksijen grubunda hafifçe yüksek olduğu gözlendi. Serum ALT ve AST değerleri, kontrol grubuna göre izofluran grubunda anlamlı bir şekilde artmış bulundu.Sonuç: Bulgular ışığında, İzofluranın hepatotoksik etkisinin olduğu fakat, bu etkinin serbest radikal metabolizması aracılığıyla oluşmadığı düşünüldü.
Abstract
Summary: Aim: To investigate whether hepatotoxic effect of isoflurane is related with the enzymatic antioxidant defence system. Materials and Methods: Fifteen guinea pigs were separated into three groups: I-control, II-isoflurane, III-oxygen. Animals in oxygen group were treated with 100 % oxygen and animals in isoflurane group with isoflurane (2% v/v) in oxygen (100 %, 2 L/min) for 30 minutes for consecutive three days. Activities ofsuperoxide dismutase (SOD), calalase (CAT) and glutathione peroxidase (GSH-Px) enzymes and levels of malondialdehyde (MDA) were measured in liver tissues of the animals. Serum AST and ALT levels were also assayed. Results: SOD and CAT activities were found to be unchanged, however GSH-Px activity was found to be decreased in isoflurane and oxygen groups compared to controls. MDA level was not changed in the isoflurane group, but found to be slightly increased in the oxygen group with respect to those of controls. Serum ALT and AST levels were significantly increased in the isoflurane group according to controls. Conclusion: Isoflurane exerts hepatotoxic effects but, it seems that those effects are not mediated through free radical metabolism.
