Lokal ileri meme karsinomunda kemoterapiye yanıtın Apoptosis ile ilişkisi

Özet
SB Onkoloji Haslanesinde tedavi olan 53 lokal ileri meme karsinomlu kadın hastanın kemoterapi öncesi biyopsi ve kemoterapi sonrası masteklomi materyalleri incelenmiştir. Mitoz sayışı, ışık mikroskopik ve "terminal deoxynucleotidyl transferase (TdT)-mediated deoxyuridine triphosphate (dUTP)-biotin nick end-labeling" (TÜNEL) yöntemiyle saptanan apoptotik indeks ve bunlarda kemoterapiye bağlı olarak meydana gelen değişiklikler araştırılmıştır. Işık mikroskopunda tümör perifer ve merkezinde ayrı ayrı değerlendirilen apoptotik hücre sayılarında artış saptanmıştır. TÜNEL yöntemiyle yapılan apoptotik hücre sayımı ve mitoz sayımlarında azalma görülmüştür. Apoptosis sayısındaki kemoterapi sonrası artış, kemolerapötiklerin apoptosisi indükliyerek etki gösterdikleri görüşü ile uyumludur. Bulgularınız apoptosis sayısındaki değişikliklerin kemoterapi etkisini değerlendirmede kullanılabilecek parametrelerden biri olabileceğin! düşündürmektedir.
Abstract
Pre-chemotherapy biopsiesand post-chemotherapy mastectomies of 53 women with locally advanced breast carcinoma who were treated at Ankara Oncology Hospital were evaluated. Mitotic count. apoptotic İndices determined by light microscopy and terminal deoxynucleotidyl transferase (TdT) mediated deoxyuridine triphosphate (dUTP)-biotin nick end-labeling (TÜNEL) technique and changes in the values of these parameters after chemotherapy were investigated. Apoptotic celi counis which was carried out on tumor periphery and center were increased. There were decreases in the apoptotic celi counis with TÜNEL technique and decreases in mitotic counis. The increase in the apoptotic counts in post-chemotherapy specimens were consistent with the idea that chemotherapeutics exert their etfects by inducing apoptosis. Qur findings suggest that changes in apoptosis count could be used as one of the parameters in the evaluation of chemotherapy effect.
